维甲酸联合低浓度化疗药物对肺腺癌A549细胞的作用

摘    要: 目的:观察全反式维甲酸和低于临床用药浓度不同剂量的化疗药物吉西他滨、顺铂等及两者联合处理A549人肺腺癌细胞株,以了解不同药物、不同剂量及不同作用时间对A549细胞增殖、细胞周期的影响。方法:用MTT法测定不同药物组相对于对照组的A549细胞增殖抑制率,并用流式细胞学方法检测各药物组及对照组的细胞周期分布。结果:维甲酸联合应用低浓度的顺铂和吉西他滨可显著抑制A549细胞的增殖,随着化疗药物的浓度的增加,抑制率也一并增加,并在达到同样增殖抑制率的效果下降低顺铂和吉西他滨的药物浓度。结论:维甲酸与低浓度化疗药物联合应用有希望显著减少药物的剂量,降低化疗药物的毒性,增强疗效。

Abstract:Objective To investigate the effects of all-trans retinoic acid,chemotherapy agents of different doses below clinic therapy concentration,e.g.gemcitabine and cisplatin,and their combination treatment on human lung adenocarcinoma cell line A549,to understand the effects of different drugs,different doses and different treatment time points on the proliferation and apoptosis of A549 cell.Methods MTT assay was used to determine the proliferation inhibition rates of different drug groups against A549 cell comparing with the control group,and the flow cytometry method was used to detect the cell cycle distribution of the drug groups and the control group.Results Retinoic acid combined with low concentrations of cisplatin and gemcitabine significantly inhibited the proliferation of A549 cells;the inhibit rate increased with increasing concentrations of chemotherapy drugs;and the concentrations of cisplatin and gemcitabine were lowered when the same inhibitory effect was achieved.Conclusion The combination therapy of all-trans retinoic acid and low concentrated chemotherapeutic drugs may have remarkable effect in reducing drug dosage and toxicity and improving efficacy.
