脑外伤小鼠海马TNF-α表达的变化

  目的 探讨脑外伤小鼠海马肿瘤坏死因子α（TNF-α）表达的变化。方法 60只Balb/c小鼠随机分为2组，假手术组（30只）和脑外伤组（30只）。每组依据手术后不同时间点再分6 h、1 d、3 d 3组，每组10只。免疫组化和Western blot检测各组小鼠海马TNF-α蛋白的表达。RT-PCR方法检测各组小鼠海马TNF-α mRNA的表达。结果 免疫组化和Western blot结果发现，脑外伤组小鼠海马TNF-α蛋白表达量明显高于假手术组（P<0.05）；RT-PCR结果也显示脑外伤组小鼠海马TNF-α mRNA表达量明显高于假手术组（P<0.05）。结论 脑外伤小鼠海马TNF-α表达明显升高。

The Expression Changes of TNF-α in Mice Hippocampus after Traumatic Brain Injury

Objective To study the expression changes of TNF-α in mice hippocampus after traumatic brain injury.Methods Sixty Balb/c mice were randomly divided into sham group(S group,n = 30) and traumatic brain injury group(T group,n = 30);each group was further divided into three subgroups at different time points(6 h,1 d and 3 d,n =8) after the surgery.TNF-α expression was determined in hippocampus of all groups by immunohistochemistry method and Western blot method;TNF-α mRNA in hippocampus of all groups was observed by RT-PCR method.Results TNF-α expression in hippocampus increased significantly higher in T group than in S group(P < 0.05),so did TNF-α mRNA expression.Conclusion The expression of TNF-α in hippocampus increased significantly in mice after traumatic brain injury.
