颌下腺导管细胞体外分离纯化的实验研究

摘    要:目的:体外分离纯化颌下腺(SMG)导管细胞.方法:选用8 d龄SD大鼠15只,无菌条件下取出颌下腺,胰蛋白酶消化,等密度梯度离心分离纯化导管细胞,免疫组织化学染色进行细胞鉴定,分光光度计检测所获得的各层细胞带上清液中淀粉酶的活性.结果:所获得的上层区带细胞免疫组织化学染色Anti-Pan cytokeratin、Anti-EMA呈阳性表达.同时淀粉酶活性检测上、下层区带分别为6%和72%.结论:应用等密度梯度离心的方法可获得纯化程度较高的SMG导管细胞.

Abstract:Objective: To isolate and purify ductal cells of submandibular glands (SMG) in vitro. Methods: The ductal cells of SD rats were isolated with density-gradient centrifugation under aseptic condition. Immunohistochemistry staining was used to determine the cells. The level of isolation and purification was evaluated by assay of amylase activity. Results: The cells in upper zone were positively stained with Anti-Pan cytokeratin and Anti-EMA,which were ductal specific antibodies. The activity of amylase of cells in upper and lower zones were 6% and 72% respectively. Conclusion: Pure ductal cells could be obtained with density-gradient centrifugation.
