Bcl-2蛋白及caspase-3基因表达与暴发性肝衰竭肝细胞凋亡关系的研究

摘    要:目的:研究Bcl-2蛋白与caspase-3基因表达在实验性暴发性肝衰竭(FHF)中对肝细胞凋亡的作用。方法:用脂多糖和D-氨基半乳糖制备FHF小鼠模型;免疫组化法检测肝组织内Bcl-2蛋白表达;原位杂交法检测肝组织内ccqspcqse-3基因表达;缺口原位末端标记技术检测肝细胞凋亡。结果:模型组用药后2h Bcl-2有较多表达,4h表达最多,以后逐渐减少,4h与2h比较差别显著(P〈0.05),与8h、12h比较差别十分显著(P〈0.01)。模型组用药后2h caspase-3开始少量表达,8h达最高峰,并出现典型的肝细胞凋亡改变。12h表达下降,且同时存在肝细胞凋亡和坏死。Bcl-2与caspase-3表达及肝细胞凋亡均呈负相关。结论:在FHF中,肝细胞凋亡与caspase-3的激活有关;Bcl-2可能通过抑制caspase-3的活性而抑制肝细胞凋亡。

Abstract:Objective: To study the effect of the expressions of Bcl-2 protein and caspase-3 gene on hepatocyte apoptosis in the experimental model of fulminant hepatic failure(FHF).Methods: The experimental mouse model of FHF was established by injecting lipopolysaccharide(LPS) and D-galactosamine(D-GalN).The expressions of Bcl-2 protein and caspase-3 gene in liver tissue were determined by immunohistochemistry method and in situ hybridization,respectively.The hepatocyte apoptosis was examined by TUNEL method.Results: In FHF model group,Bcl-2 protein expression significantly increased 2 hours after the injection,reached the peak 4 hours later,and then gradually decreased.Bcl-2 protein expression 4 hours after the injection was significantly higher than that 2,8,and 12 hours after the injection,respectively(P < 0.05,P < 0.01,P < 0.01,respectively).Caspase-3 gene expression was found 2 hours after the injection of LPS and D-GalN,reached the peak 8 hours after the injection,and decreased 12 hours later.The typical apoptotic hepatocytes significantly increased 8 hours after the injection,and the hepatocyte apoptosis and hepatic necrosis appeared simultaneously 12 hours later.Bcl-2 protein expression negatively correlated with both the hepatocyte apoptosis and caspase-3 gene expression.Conclusion: The hepatocyte apoptosis is correlated with the activation of caspase-3 in FHF,and Bcl-2 may inhibit the hepatocyte apoptosis by inhibiting the activation of caspase-3 expression.
