滨蒿内酯对豚鼠哮喘模型的炎性介质抑制作用

摘    要:目的:研究滨蒿内酯(Seo)对豚鼠哮喘的抗炎作用及机制。方法:制备迟发性哮喘豚鼠模型,免疫组化法检测豚鼠血浆和支气管肺泡灌洗液(BALF)中白介素5(IL-5)含量;电镜法对豚鼠肺组织细胞溶酶体磷脂酶A:(PLA2)进行染色,井通过图像分析定量检测其含量。结果:哮喘组IL-5及PLA2的含量与正常对照组比较明显增高(P〈0.05);提前雾化给予42mg/L,340mg/L的Seo组与哮喘模型组比较明显降低IL-5及PLA2含量(P〈0.05)。结论:Seo可能通过抑制IL-5及PLA2发挥其对支气管哮喘的作用。

Abstract:Objective: To study the anti-inflammatory effect of scoparone(Sco) on asthma and its mechanism in guinea pigs.Methods: Guinea pig model of late asthmatic reaction(LAR) was established.The content of interleukin-5(IL-5) in serum and bronchial alveolar lavage fluid(BALF) was detected by using immunohistochemical method.The content of phospholipase A_(2)(PLA_(2)) in the lysosomes of pulmonary tissues was detected by image analysis after staining.Results: The contents of IL-5 in serum and BALF and PLA_(2)in the lysosomes of pulmonary tissues in the asthma group were significantly higher than those in the control group(P<0.05).The contents of IL-5 and PLA_(2)in the treatment group(Sco of 42 and 340 mg/L) were significantly lower than those in the asthma group(P<0.05).Conclusion: Sco can inhibit inflammation by lowering the contents of IL-5 in serum and BALF and PLA_(2)in pulmonary tissues in asthmatic guinea pig model.
