滨蒿内酯对哮喘豚鼠脾淋巴细胞转化和IL-2生成的影响

摘    要:目的:研究哮喘豚鼠脾淋巴细胞转化和白细胞介素2(IL-2)含量的变化及滨蒿内酯(Sco)对其的影响。方法:建立卵蛋白致敏的哮喘豚鼠模型。MTT比色分析法测定光密度值,比较不同浓度Sco对淋巴细胞增殖、植物血凝素(PHA)诱导的T淋巴细胞转化以及脾细胞分泌IL-2活性的影响。结果:Seo剂量依赖地对哮喘豚鼠淋巴细胞增殖有抑制作用;Seo能够抑制PHA诱导的哮喘朦鼠T淋巴细胞转化;哮喘豚鼠IL-2生成水平高于正常对照组。Seo也可抑制哮喘豚鼠IL-2的活性。结论:Seo在体外抑制哮喘豚鼠脾淋巴细胞转化和脾淋巴细胞产生IL-2的水平,可能是其治疗哮喘的免疫学机制之一。

Abstract:Objective: To study the effects of scoparone(Sco) on splenic lymphocyte transfromation and interleukin-2(IL-2) production in asthmatic guinea pigs in vitro.Methods: Asthmatic guinea pig model was established.The optical density value was measured by using MTT method.The effects of different concentrations of Sco on lymphocyte proliferation,phytohemagglutinin(PHA)-induced T lymphocyte transformation,and IL-2 production were compared.Results: Sco inhibited lymphocyte proliferation in a dose-dependent manner and PHA-induced T lymphocyte transformation.The IL-2 production in asthmatic guinea pigs was higher than that in normal controls.Sco also inhibited IL-2 activity.Conclusion: Sco could inhibit splenic lymphocytes transformation and IL-2 production in asthmatic guinea pigs in vitro,which may be one of the immunological mechanisms in treating asthma.
