兔骨髓间充质干细胞的分离、培养和鉴定

目的探讨兔骨髓间充质干细胞分离、培养和鉴定的方法,比较与人骨髓间充质干细胞表面抗原表达的差异。方法梯度离心法提取兔骨髓中的间充质干细胞,常规贴壁生长,免疫组化和流式细胞的方法检测细胞CD44、CD29、CD34的变化。结果培养的细胞呈纤维状、克隆样生长;免疫组化表明CD29、CD44呈阳性表达,CD34呈阴性表达,流式细胞的结果表明90.26%的细胞表达CD29,89.76%的细胞表达CD44,仅有5.15%的细胞表达CD34。结论兔骨髓间充质干细胞表面抗原的部分表达与人相同,可用于相关的基础研究。

Isolation、Culture and Identification of Rabbit Mesenchymal Stem Cells

Objective To evaluate the methods of isolation,culture and identification of rabbit mesenchymal stem cells (MSCs),and compare the expression of cell-surface markers between rabbit and human. Methods Rabbit MSCs were obtained from the femurs of Adult New Zealand White rabbits by centrifugation and analyzed by Immunofluorescence and flow cytometry to measure the changes of CD29,CD44 and CD34. Results Most cell lines were composed of cells with a characteristic spindle shape. Immunocytochemistry revealed that Rb-MSCs were positive in CD44 and CD29 and negative in CD34. Flow cytometric analysis confirmed that 90.26% of the cells expressed CD29 and 89.76% of the cells expressed CD44; only 5.15% of the cells expressed CD34 in cultured cells. Conclusion The cell-surface markers in rabbit MSCs are partly the same as those in human MSCs,so the rabbit MSCs can be used in researches instead of human MSCs.
