摘    要:采用新鲜肺癌组织制成的细胞悬液及肺腺癌细胞株做为免疫原,免疫Balb/c小鼠。取脾细胞与经活体增殖的无支原体污染的高活力SP2/O骨髓瘤细胞融合,联合应用酶联免疫吸附和多组织切片免疫组化染色法进行筛选,得到两株分泌对肺癌具高特异性的单克隆抗体的杂交瘤细胞株。抗体与被检测的大多数人体正常组织无交叉反应,可初步用于肺癌病理鉴别诊断。

Abstract:Two murine monoclonal antibodies, 3D5 and 1F11, against human lungcarcinomas were generated by immunizing with fresh human lung carcinomacells and lung adenocarcinoma cell line, AGZY-83-a. The antibody 3D5 reactedwith lung tumor tissues (64 of 101 cases positive), especially with lung squa-mous carcinoma (32 of 35 cases positive) by using AEC immunoperoxidasetechnique. The results showed the heterogenicity in binding some lung tumorcells and the antigens were the lung carcinome associated antigens. Usingmycoplasma-free and high cell viability myeloma cells is very important for cellfusion and clonal growth. Combination of ELISA with immunohistochemicalstaining of multiple tissue embeded block sections is a good method for monoclonalantibody screening.
