摘    要:目的 :以钳夹技术评估高脂膳食建立胰岛素抵抗 (IR)动物模型的可行性。方法 :以脂肪热比为 5 9%(其中猪油占 39% ) ,蛋白质热比为 2 1% (其中酪蛋白占 31% ) ,碳水化合物热比为 2 0 % (其中玉米淀粉占 30 % )的特殊高脂膳食 ,喂养 4周后建立SD大鼠钳夹技术。结果 :高脂膳食喂养大鼠 4周后 ,葡萄糖输注率 (GIR)显著低于正常膳食喂养大鼠 (P <0 .0 1) ,且GIR水平与大鼠体重显著负相关 (γ =- 0 .75 2 ,P <0 .0 1)。结论 :建立SD大鼠的钳夹技术各项指标可靠、准确、稳定 ;高脂膳食喂养 4周后 ,钳夹技术证明大鼠胰岛素敏感性下降 ,提示复制IR大鼠模型成功。

Abstract:Objective: Our aims were to establish the animal model of insulin resistance (IR) with high fatty diet and to evaluate it by using hyperinsulinemic euglycemic clamp technique. Methods: Adult SD male rats were fed with high fatty diet(59% of calories fat, 21% of calories protein, 20% of calories carbohydrate) for 4 weeks and then evaluated by using hyperinsulinemic euglycemic clamp technique. Results: After the SD rats were fed with high fatty diet for 4 weeks, the average body weight of the rats fed with high fatty diet were greatly higher than that of the control rats. The core marker of clamp forceps technique, glucose infusion rate (GIR), was obviously lower in rats with high fatty diet than that in rats fed with normal diet (P<0.01). And significant negative correlation could be observed between GIR and the weight of rats (r=-0.752, P<0.01). Conclusion: Clamp forceps technique was used for the first time in the evaluation of animal model of IR created by using high fatty diet and such animal model could also be used in the research for IR related diseases of human beings.
