长白山白眉蝮蛇类凝血酶基因特异片段的制备与分析

摘    要:目的:分离和纯化长白山白眉蝮蛇类凝血酶cDNA片段,为克隆全长类凝血酶cDNA奠定基础。方法:应用RT-PCR方法获得长白山白眉蝮蛇类凝血酶cDNA片段,克隆于pGEM-T载体上,进行测序和NCBI数据库同源性分析。结果:获得长白山白眉蝮蛇类凝血酶cDNA片段长度为289bp,该片段与其他种蝮蛇类凝血酶DNA片段有同源性。结论:本实验克隆了长白山白眉蝮蛇类凝血酶基因片段,根据不同种属蛋白质家族成员的保守性和同源性设计引物可以有效地克隆家族成同的DNA片段。

Abstract:Objective: This study was to clone the fragment of thrombin-like enzyme (TLE) from A.H.Ussuriensis Emelianov. Methods:Fragment of TLE cDNA was amplified by RT-PCR,then cloned into pGEM-T vector and sequenced.Results:The 289 bp fragment of TLE cDNA was cloned.Conclusion:We have cloned the part of TLE cDNA from A.H.Ussuriensis Emelianov.The result will give us a firm foundation to clone the full length TLE cDNA.
