摘    要:目的:探讨成年大鼠骨髓间充质干细胞(MSC)体外分化为神经前体细胞的可能性.方法:MSC原代培养、传代后在细胞因子和氧化剂作用下进行诱导.形态学观察,免疫细胞化学、流式细胞分析和细胞电生理检查.结果:MSC体外可诱导为神经前体细胞,诱导后有表面抗原nestin表达,5 h分化率49.8%,同时具有早期神经干细胞电流特性.结论:骨髓来源的神经前体细胞治疗中枢神经系统疾病成为可能.

Abstract:Objective: To explore the possibility of transdifferentiation of bone marrow mesenchymal stem cells (MSC) of adult rat to neural precursor cells in vitro. Methods: After the primary culture and subculture differentiation of MSC was induced into neural precursor cells by cytokine and oxidane . Gross observation, immunocytochemistry analysis, flow cytometry analysis, and electrophysiological examination were performed. Results: The MSC could be transdifferentiated to neural precursor cell with the expression of Nestin. The inducible rate of MSC was 49.8%. The induced cells showed different electrophysiological features from the MSC,but similar to the neural stem cells. Conclusion: Neural precursor cells derived from bone marrow have the possibility in the therapy of central nervous system damage.
