蟾蜍毒素粗提物对白血病K562细胞的杀伤作用

摘    要:目的:观察蟾蜍毒素提取物对白血病K562细胞杀伤作用及对细胞周期的影响。方法:用无水乙醇、二甲基亚砜(DMSO)、PBS缓冲液浸溶中华大蟾蜍的耳后腺及皮下腺分泌物,采用台盼蓝拒染法检测细胞增殖抑制作用,流式细胞仪检测对细胞周期的影响。结果:3种浸液均能抑制K562细胞的增殖,其抑制强度依次为乙醇浸液>DMSO浸液>PBS浸液。5 ng/ml的乙醇浸液、50 ng/ml的DMSO浸液、500 ng/ml的PBS浸液均能使K562细胞发生G2/M期阻滞。结论:蟾蜍毒素粗提物对白血病K562细胞增殖具有明显的抑制作用,并伴有G2/M期阻滞。

Abstract:Objective: Our aim was to determine the effect of toad toxication on proliferation and progression of cell cycle in cultured leukemia K562 cells. Methods:The secretion of sebaceous gland of Chinese toad was soaked with etha-nol, DMSO, and PBS. Three kinds of crude extraction (extraction E, D, and P)were made. Inhibition of cell proliferation was examined by typanblue dye exclusion method, and progression of cell cycle was analyzed by flow cytometry in K562 cells. Results:Proliferation of K562 cells was obviously inhibited by 3 kinds of crude extraction. The order of strength of inhibitory effect of these crude extraction was extraction E, extraction D, extraction P (from the highest to the lowest). The G2/M phase arrest of cell cycle was significantly induced by extraction E (5 ng/ml) , extraction D (50 ng/ml) , and extraction P (500 ng/ml) . Conclusion: Proliferation of K562 cells was inhibited by crude extraction, and inhibitory effect of cell proliferation was accompanied with C2/M arrest of cell cycle.
