摘    要:目的探讨2,3吲哚醌(isatin,ISA)对MPTP制备的C57BL/6J小鼠帕金森病(PD)模型黑质中BAX基因表达影响。方法 C57BL/6J小鼠随机分为五组,空白对照组,MPTP损伤组,ISA低(50 mg/kg,ig)、中(100 mg/kg,ig)、高剂量(200 mg/kg,ig)预处理+MPTP组。结果与对照组相比,MPTP损伤的小鼠黑质中BAX免疫反应阳性表达增加(P〈0.01);在ISA 100 mg/kg和200 mg/kg预处理组与MPTP损伤组相比,黑质中BAX免疫反应阳性表达显著降低(P〈0.05)。结论表明ISA对MPTP造成的PD小鼠有明显的保护作用。

Abstract:Objective:To explore the effect of isatin on substantia nigra(SN) BAX gene expression in MPTP-induced PD model in mice.Methods:C57BL/6J mice were divided into 5 groups:① control group;② MPTP group;③ ISA 50 mg/kg ig + MPTP group;④ ISA 100 mg/kg ig + MPTP group;and ⑤ ISA 200 mg/kg ig + MPTP group.Then immunohistochemical techniques was used to observe the BAX gene’s expression in substantia nigra in each mouse.Results:The results showed that compared with the control group,the BAX immunoreactive expression in SN was higher in MPTP injury mice(P<0.01);in ISA 100 mg / kg and 200 mg / kg pretreatment group,the BAX immunoreactive expression in SN was significantly reduced compared with the MPTP injury group(P<0.05).Conclusion:It is shown that the ISA has the protective effect on MPTP-induced PD model in mice.
