疏肝健脾方对乳腺癌组织淋巴管生成及Ang-2、nm23表达的影响

摘    要:目的观察中药疏肝健脾方对乳腺癌组织淋巴管生成及Ang-2、nm23表达的影响,探讨复方中药抑制乳腺癌淋巴转移机制。方法 50例乳腺癌病人随机分成2组,疏肝健脾方组(实验组)30例,对照组20例,两组病人经巴德活检针活检,标本行组织学和免疫组化法检测乳腺癌组织微淋巴管密度和Ang-2、nm23的表达,然后给予CTF方案化疗2个周期,实验组同时口服疏肝健脾方制剂,化疗结束后行乳腺癌手术,手术标本检测微淋巴管密度和Ang-2、nm23的表达。比较两组标本微淋巴管密度和Ang-2、nm23的表达的差异。结果实验组和对照组化疗后肿瘤体积均明显缩小,淋巴管密度明显减少,两组比较有显著性差异(P<0.05或P<0.01);实验组和对照组乳腺癌组织中化疗前Ang-2阳性表达率无显著性差异(P>0.05),化疗后Ang-2阳性表达率均明显降低,组间比较有显著性差异(P<0.05);实验组和对照组乳腺癌组织中化疗前nm-23阳性表达率无显著性差异(P>0.05),化疗后nm-23阳性表达率均有所升高,两组化疗后nm-23阳性表达率有显著性差异(P<0.05)。结论疏肝健脾方可能通过干预Ang-2、nm23的表达,影响的淋巴管生成抑制乳腺癌淋巴转移。

Abstract:Objective: To observe effects of compound Chinese medicine Shugan jianpi formula on lymphangiogenesis and expressions of Ang-2 and nm23 in breast cancer tissue and to explore the mechanism of tumor metastasis resistance.Methods:Fifty breast cancer patients were divided into the two groups randomly,30 cases in the Shugan jianpi formula group(test group) and 20 cases in the control group.All the patients were given CTF program chemotherapy,and the test group were given Shugan jianpi formula at the same time.After two cycles of chemotherapy,all the patients were given operations,and then micro lymphatic density and Ang-2 and nm23 expressions were detected in specimens.Results:The tumor volume and lymphatic density were decreased apparently after chemotherapy in both of the groups,with significant differences between the two groups(P<0.05 or P<0.01).Ang-2 positive expression rate in breast cancer tissue before chemotherapy had no significant difference between the two groups(P>0.05),but decreased apparently after chemotherapy in both the groups,with significant differences between the two groups(P<0.05).The expression rate of nm-23 before chemotherapy had no significant difference between the two groups(P>0.05),but increased apparently after chemotherapy in both of the groups,with significant differences between the two groups(P<0.05).Conclusion: Shugan jianpi formula may influence the lymphangiogenesis and inhibit breast cancer lymph node metastases through intervening the expressions of Ang-2 and nm23.
