灵草液对Alzheimer病模型大鼠脑组织NO、NOS的影响

摘    要:目的:观察灵草液对AD模型大鼠脑组织中一氧化氮、一氧化氮合酶的影响。方法:SD健康雄性大鼠,随机分成7组,采用大鼠脑组织立体定位微量注射技术,用IBO损毁大鼠双侧nbM,建立AD模型大鼠。手术后灵草液连续灌胃共20d,1d1次,每次2m l。灌胃期满后即将大鼠断头处死,并立即在冰盘中开颅取脑做组织匀浆,取其上清液测定各生化指标。结果:用IBO损毁大鼠双侧nbM,建立的AD模型大鼠脑组织一氧化氮(NO),一氧化氮合酶(NOS)均明显升高。灵草液能显著地降低AD模型大鼠脑组织NO含量和NOS的活性,与模型组比较有显著性差异(P<0.01,P<0.05)。结论:灵草液能降低AD模型大鼠的NO含量和NOS的活性。

Abstract:Objective: To observe the effects of lingcao liquid on the brain nitric oxide(NO)、 nitric oxide synthase(NOS)in the brain tissue of the rats model with non-hereditary Alzheimer′s disease.Methods: Healthy male SD rats(8weeks~12weeks,170g~230g) were randomly divided into 7 groups.In the research,AD animal model is established by injecting IBO 1μl(5μg) into rats brain to damage bilateral nbM in rats brain.Group Ⅰwas normal group;groupⅡ was AD animal model;group Ⅲ,Ⅳ,Ⅴ,Ⅵ,Ⅶ were AD animal model + orally normal saline,Naofukang 0.3g/kg·d,lower dosages of Lingcao liquid,medium dosages Lingcao liquid,high dosages of Lingcao liquid were perfused into the stomach respectively for 20 days,2ml,once per day,while the normal control group and model group rats had not been treated.After that,AD rats were cut off the head to take out the brain tissue in ice tray and the levels of NO and NOS in rats brain were analyzed by biochemical method.Results: Rats'bilabial nbM was damaged by IBO,NO and NOS in model group brain increased significantly(P<0.01);NO、NOS of brain of normal saline group didn′t change significantly(P>0.05);NO and NOS of brain of Naofukang group and Lingcao liquid groups decreased significantly(P<0.01).The significant difference could seen compared with the model group.Conclusion: Lingcao liquid can decrease significantly the levels of NO and NOS in brain of AD model group.
