TRAIL、Survivin mRNA表达与胃癌凋亡发生的相关关系

目的:检测肿瘤坏死因子相关凋亡诱导配体(TRAIL)和SurvivinmRNA在胃癌中的表达及其与凋亡的相关关系。方法:利用原位杂交技术检测67例胃腺癌及对应的正常胃粘膜中TRAIL和SurvivinmRNA的表达,同时利用原位末端标记技术(TUNEL)检测癌组织中的凋亡指数(AI)。结果:①TRAILmRNA在正常胃粘膜中的阳性表达率高于癌组织中的阳性表达率。②胃癌组织中细胞凋亡指数为1.85+0.66。③TRAILmRNA阳性组与阴性组、SurvivinmRNA阳性组与阴性中凋亡指数分别存在着显著性差异(P<0.05)。④TRAILmRNA和SurvivinmRNA在胃癌中的阳性表达率呈明显的负相关(P<0.05)。⑤TRAILmRNA和SurvivinmRNA与胃癌组织中的患者的年龄、性别、肿瘤直径、部位、大体分型、侵润深度及有无淋巴结转移无明显的关系(P>0.05),与肿瘤的分化程度有关(P<0.05)。结论:TRAIL和Survivin在胃癌凋亡过程中起着重要的作用,TRAIL可诱导凋亡,Survivin则抑制凋亡发生。

Study on the Relationship Between the TRAIL ,Survivin and Gastric Carcinoma Apoptosis

Objective: To detect the expression of TNF related apoptosis induced by ligand and Survivin in gastric carcinoma, and their association with tumor cell apoptosis. Methods: By using in situ hybridization technology to detect the expression of TRAIL mRNA and Survivin mRNA in 67 cases of gastric carcinoma and normal mucosa, at the same time to detect the apoptosis index (AI) in cancer tissues by using TUNEL technology.Results: ① The positive rate of TRAIL mRNA in normal mucosa was higher than it in cancer tissues; Survivin mRNA was only expressed in carcinoma. ② The AI was 1.85+0.66 in gastric carcinoma tissues. ③ It was statistically significant of AI between the TRAIL mRNA,Survivin mRNA positive group and negative group (P<0.05). ④ The expression of survivin mRNA in carcinoma was converse relationship to that of TRAIL (P<0.05) .⑤ It had no statistically significant between TRAIL、Survivin mRNA and tumors diameter , site, gross classification , invasion depth , lymph nodes metastasis and the patients′ ages and sexes(P>0.05),and it had significant relationship with tumours differentiation (P<0.05). Conclusion: TRAIL can induce apoptosis and Survivin can depress the happening apaptosis.
