溃结宁合剂对溃疡性结肠炎大鼠IL-1β和IL-4的影响

摘    要:目的:观察溃结宁合剂对溃疡性结肠炎大鼠结肠黏膜病理组织学变化和对IL-1β和IL-4表达的影响。方法:将41只SD大鼠随机分为正常组、模型组和溃结宁低、中、高剂量组,用2,4-二硝基氯苯和乙酸制备UC大鼠模型。给药后第15d处死动物,取各组所得结肠标本,行HE染色,观察各组病理组织学变化,并用免疫组化法染色,观察各组IL-1β和IL-4表达的变化。结果:溃结宁各组均可明显改善UC大鼠结肠黏膜的病理组织学损伤改变,并能明显降低UC大鼠结肠黏膜IL-1β的表达和提高IL-4的表达。结论:溃结宁可能通过增加IL-4的释放和抑制IL-1β的释放而治疗UC,促进结肠黏膜恢复。

Abstract:Objective:To observe the effect of Kuijiening decoction(KJN)on the expression of IL-1β and IL-4 of colonic mucosa of rats with ulcerative colitis(UC). Methods: 41 SD rats were randomized into normal group, model group, KJN treatment group 1(low dosage), group 2 (medium dosage)and group 3(high dosage). The model rats were induced by dinitrochlorobenzene and acetic acid. All rats were killed and the colonic mucosa samples were taken after 15 days' treatment. The colonic mucosa pathological changes, IL-1β and IL-4 of colonic mucosal tissues were observed by HE staining and immunohisto-chemistry staining. Results: Compared with model group, the colonic mucosa pathological lesions of KJN treatment group were obviously alleviated and the level of IL-1β in colonic mucosa was decreased, the level of IL-4 was increased significantly. Conclusion: KJN can regulate the expression of IL-1β and IL-4 in colonic mucosa, which may contribute to its action in reducing the colonic mucosa of UC.
