纳气平喘颗粒对大鼠哮喘肺组织病理变化影响探讨

摘    要:目的探讨纳气平喘颗粒对哮喘大鼠呼吸道病理学变化的影响及其对肺水代谢的药理作用。方法采用卵清蛋白(Ovalbumin,OVA)致敏方法建立支气管哮喘大鼠模型。Wistar大鼠69只随机分为正常对照组、哮喘模型组、纳气平喘颗粒干预组、地塞米松干预组。观察和比较各组大鼠肺内各段气道的病理学变化,测定肺组织湿干重比值(Wet Dry ratio,W/D),并进行炎性细胞和杯状细胞计数。结果哮喘模型组与正常对照组相比,哮喘模型组呼吸道管腔狭窄,腔内有大量黏液栓(Mucous Plug)形成;黏膜上皮受损、气道平滑肌增厚、黏膜水肿(Muco-sal Edema)及炎性细胞浸润等病理学变化显著。地塞米松干预组及纳气平喘颗粒干预组上述病理学变化减轻;纳气平喘颗粒组肺组织W/D值、炎性细胞和杯状细胞数值均低于哮喘模型组,差异有统计学意义(P<0.05)。结论纳气平喘颗粒药理作用与地塞米松相似,能够有效地改善哮喘大鼠呼吸道病理学变化,对哮喘引起的黏液高分泌及黏膜水肿有一定的调节作用。

Abstract:Objective To investigate the effect of Na Qi Ping Chuan Granules on mucus hypersecretion and mucosal edema in airway of Asthmatic Rats.Methods Sixty-nine Wistar rats were divided into four groups randomly,they were asthma group,normal control group,Na Qi Ping chuan granules treated group and Dexamethasone treated group.The asthma models were established by sensitization with ovalbumin(OVA) and aluminum hydroxide on the 1 st day,15 th day,and challenged with OVA on the 25 th to 29 th days.Each group′s histopathology chances of airway were observed,lung wet-dry weight proportionality(W/D) was measured,inflammatory cells and goblet cells were counted.Results Compared with normal group,the asthma group except the chronic inflammation,also displays of serious mucous plug,heavy mucosal edema,and goblet cell hyperplasia in the respiratory tract.The inflammation changes were significantly improved in both Naqi and Dexamethasone treatment groups.The lung W/D ratio and goblet cell quantity in drug treatment groups were significantly lower than that in model group(P<0.05).Conclusion Na Qi Ping Chuan Granules have a anti-inflammatory activity,and may have potential therapeutic roles in mucus hypersecretion and mucosal edema in asthma.
