| 摘 要： | 目的:尝试提取人体肠道菌群总DNA,为研究肠道菌群结构提供依据。方法:以人体粪便为样本,使用PBS多次清洗,离心样品,沉淀粪便中的菌体。使用裂解液、溶菌酶和蛋白酶K等裂解菌体,用酚/氯仿方法抽提DNA,以提取到的肠道细菌总DNA为模板进行ERIC-PCR扩增。结果:电泳显示样品DNA条带明亮,少数样品DNA存在降解现象;ERIC-PCR扩增得到较清晰的指纹图谱。结论:经改进后的方法操作简单、实用、经济,适用于肠道菌群结构的研究。

| Abstract: | Objective: Extracting total DNA of intestinal tract bacteria from human faeces to investigate intestinal microflora.Methods: Taking the human faeces as samples,to precipitate bacteria through PBS cleaning and centrifuge.Nucleinic acid was released after the bacteria were lysed with lysate,antalzyme and proteinase K.DNA was purified with phenol/chloroform,subsequently used as templates in ERIC-PCR.Results: The electrophoresis displaied the banding-patterns of different samples were bright,minor samples were degraded,and DNA fingerprints were obtained.Conclusion: This method is simple,utility,economical and may be used to study the structural features of intestinal communities. |
