糖尿病肾病大鼠蛋白尿和肾组织WT1表达的相关性研究

摘    要:目的 观察WT1在糖尿病肾病大鼠模型中的表达,从而探讨WT1与蛋白尿的相关性及其在蛋白尿发生发展过程中的作用.方法 56只雄性Wistar大鼠,随机分为对照组和模型组,建立糖尿病肾病大鼠模型,在第1、2、4、8、12周,应用HE染色光镜观察肾小球、肾小管组织结构变化,免疫组织化学技术检测肾组织WT1蛋白的表达,二辛可宁酸(BCA)法测定24 h尿蛋白排泄量,Western bolt法检测大鼠肾皮质中WT1的表达.结果 随着模型组大鼠蛋白尿排泄量的增加,肾组织病理改变逐渐加重,WT1表达量逐渐增加,模型组大鼠的WT1表达量均高于对照组(P<0.05).WT1的表达量和24h尿蛋白排泄量之间为正相关(r=0.723 52,P<0.01).结论 WT1的表达量随着尿蛋白量的增加而逐渐增多,二者呈正相关;WT1有可能作为判定早期肾脏损伤的参考指标.

Abstract:Objective To observe the expression of WT1 in the rat model of diabetic nephropathy,so as to explore the relevance of WT1 with albuminuria and proteinuria arise in the role of the development process.Methods 56 male Wistar rats were divided into control group and model group randomly,the model of rat diabetic nephropathy was established,the diversification in organizational structure of glomerular and tubular with HE dyeing was observed in the 1,2,4,8,12 week,the expression of WT1 protein in kidney with immunohistochemical was detected,24-hour urinary protein excretion was determined with BCA method,the expression in renal cortex of rat model was detected with Western bolt method.Results With the increased of proteinuria excretion in model rats,the pathological changes of kidney tissue deteriorated gradually,the expression of WT1 increased gradually,the expression of WT1 in model rats was higher than control group (P < 0.05).The relationship between the expression of WT1 and 24-hour urinary protein excretion was positive (r =0.723 52,P < 0.01).Conclusion The expression of WT1 protein increased with the urinary protein excretion increase gradually,they have positive correlation; it may be used as an indicator to determine the early kidney damage.
