苦碟子对高同型半胱氨酸血症细胞凋亡的影响

摘    要:目的通过大鼠灌胃给予蛋氨酸复制高同型半胱氨酸血症动物模型,观察苦碟子对高同型半胱氨酸血症大鼠动脉病理变化和细胞凋亡的影响。方法将雌性Wistar大鼠共28只随机分为模型组(n=8)、空白对照组(n=7)、叶酸+VitB12组(n=7)和苦碟子组(n=6)。除空白对照组外,其他组每天2次灌胃给予蛋氨酸0.75kg,连续10周,复制高同型半胱氨酸血症动物模型。叶酸+VitB12组和苦碟子组同时给予叶酸+VitB12和苦碟子干预。检测各组大鼠血浆同型半胱氨酸、Caspase-3含量及动脉病理变化和平滑肌凋亡细胞计数。结果模型组大鼠灌胃给予蛋氨酸后血浆同型半胱氨酸、Caspase-3含量和平滑肌凋亡细胞计数均较空白对照组升高(均P〈0.01);苦碟子组血浆Caspase-3含量和平滑肌凋亡细胞计数与模型组比较降低(均P〈0.01)。结论高蛋氨酸可诱发高同型半胱氨酸血症,引发动脉粥样硬化。苦碟子对高同型半胱氨酸血症诱发的动脉粥样硬化具有预防作用.其机制与抑制细胞凋亡相关。

Abstract:Objective To establish animal model of hyperhomocysteinemia by intragastric administration of methionine in rats, in order to observe the effect of Kudiezi on arterial pathological changes and cell apoptosis in rats with hyper- homocystinemia. Methods Female Wistar rats (n = 28) were randomly divided into the model group (n = 8), blank control group (n = 7), folic acid plus VitB12 group (n = 7) and Kudiezi group (n = 6). The rats were given 0.75 g/kg methionine 2 times a day for 10 consecutive weeks by gavages to establish hyperhomocysteinemia animal model, be- sides the blank control group. Folic acid plus VitB12 group and Kudiezi group were given folic acid plus VitB12 and Kudiezi intervention respectively. Plasma content of homocysteine and Caspase-3, arterial pathological changes and smooth muscle apoptosis cell count of all the groups were detected. Results Compared with the blank control group, plasma homocysteine, Caspase-3 content and smooth muscle apoptosis cell count of the model group were significantly elevated (P 〈 0.01). Compared with the model group, plasma Caspase-3 content and smooth muscle apoptosis cell count of the Kudiezi intervention group significantly decreased (all P 〈 0.01). Conclusion High methionine can induce high hyperhomocysteinemia and further cause atherosclerosis. Kudiezi has a preventive effect for atherosclerosis in- duced by high hyperhomocysteinemia, the mechanism may be related to the inhibition of apoptosis.
