DOG1蛋白在直肠癌中的表达及其意义

摘    要:目的观察DOG1蛋白存直肠癌中的表达,探讨其临床其意义。方法选取30例确诊为直肠癌且以全直肠系膜切除原则行直肠癌根治术的患者,取肿瘤组织及瘤旁间质组织进行常规包埋切片,行免疫组织化学染色,进行DOG1、CD117检测,观察分析检测结果.同时对DOG1阳性表达与患者年龄、性别、肿瘤直径、细胞核异形性等的关系进行分析.随访患者3年,观察DOG1阳性表达与患者生存率的关系。结果肿瘤组织中DOG1蛋白多呈弥漫性表达,肿瘤组织DOGI阳性表达率为23.33%.瘤旁间质组织DOGI阳性表达率为10.00%,肿瘤组织DOG阳性表达率明显高于瘤旁间质组织,差异有统计学意义(P〈0.05);肿瘤组织CD117阳性表达率为80.00%,瘤旁间质组织DOG1阳性表达率为33.33%,肿瘤组织CD117阳性表达率明显高于瘤旁间质组织,差异有统计学意义(P〈0.05);DOG1蛋白阳性表达与肿瘤细胞丰富程度及细胞核异形性有关,差异有统计学意义(P〈0.05);DOG1表达阳性者1年生存率与表达阴性者差异无统计学意义(P〉0.05),但3年生存率明显低于DOG1表达阴性者,差异有统计学意义(P〈0.05),结论DOG1蛋白在直肠癌患者肿瘤组织中表达高于瘤旁间质组织,且阳性表达率与肿瘤细胞丰富程度及核异彤性程度有父,而且其阳性表达与患者预后有关,临床应引起重视。

Abstract:Objective To observe the expression of DOG1 protein in rectal cancer, to explore its clinical significance. Methods 30 cases diagnosed as rectal cancer and carried out with total mesorectal excision surgery were collected, the tumor tissue and peritumoral stromal tissue were detected by the conventional embedded sections and immunohistoehemistry staining, DOGI and CDl17 detection were carried out to analyze the result. The relationship between the DOG1 positive expression and the age, gender, tumor size, nuclear atypia were analyzed. Patients were followed for 3 years to observe the relationship of the DOGl-positive expression and survival rate. Results Diffuse expression of DOGI protein in tumor tissue was found, positive expression rate of DOG1 in tumor tissue was 23.33%, positive expression rate of DOG1 in peritumoral stromal tissue was 10.00%, positive expression rate of DOG1 in tumor tissue was higher than that in peritnmoral stromal tissue, the difference was statistically significant (P 〈 0.05); positive expression rate of CD117 in tumor tissue was 80.00%, positive expression rate of CD117 in peritumoral stromal tissue was 33.33%, positive expression rate of CD117 in tumor tissue was higher than that in peritumoral stromal tissue, the difference was statistically significant (P 〈 0.05); richness of the tumor cells anti nuclear atypia were related to DOGI protein positive expression, the differences were statistically signifi- cant (P 〈 0.05); the difference of I-year survival rate between the positive anti negative expression of DOGI was not statislit:ally significant (P 〉 0.05); but the 3-year survival role in patients with positive expressinn of DOGI was lower than that in palienls wilh negative expression, the difference was statistically significant (P 〈 0.05). Conclusion DOG I protein expression in tmnor tissue of patients with rectal cancer is much higher than that in peritmnoral stromal tissue, anti it relate to richness of the tumor ceils anti nuclear alypia; its pnsitive, expression is closely related to clinical prognosis, the clinic should pay more allention.
