疏肝通络强精方对改进型实验性精索静脉曲涨大鼠睾丸EGF、TGF-β1表达的影响

摘    要:目的从分子水平探讨疏肝通络强精方治疗精索静脉曲张致不育的作用机制。方法选择青春期清洁级雄性SD大鼠50只,按随机数字表法分为假手术组、模型组、手术治疗组、桃红四物合五子衍宗汤组(桃红合五子绀)、疏肝通络强精方组,每组各10只。1个月后,对各组均按1mL/100g体重灌胃,给药或生理盐水.每日1次,普食喂养,自南饮食饮水,连续60d后,处死并取出睾丸,免疫组化法检测睾丸组织转化生长㈥子(TGF—β1)及表皮生长因子(EGF)的表达、结果①模型组睾丸组织EGF表达量较各治疗组明显减少,差异有统计学意义(P〈0.05),(2)模型组睾丸组织TGF—β1表达量较各瀹疗组明显增多,差异有统计学意义(P〈0.05或P〈0.01)。结论文验性精索静脉曲张青春期大鼠睾丸局部生长因子EGF、TGF—β1的变化是影响生精功能并导致精子质苗下降的可能机制之一;疏肝通络强精方能明显下调TGF—β1表达,并能上调EGF的表达,从而提高精液质量.可能是疏肝通络强精方埘改进型实验性精索静脉曲张青春期大鼠睾丸生精功能保护作用的凋控机制之一。

Abstract:Objective To discuss the mechanism of Shugan Tongluo Qiangjing Decoction in the treatment of infertility caused by varicocele from molecular level. Methods 50 clean male SD rats during puberty were chosen, and divided into sham operation group, model group, surgical treatment group, Taohong Siwu and Wuzi Yanzong Decoction group (Taohong and Wuzi group), Shugan Tongluo Qiangjing Decoction group according to random number table, with 10 cases in each group. After one month, the medicine was given to all groups in 1 mL/100 g weight. All groups were given medicine or normal saline, once a clay, and fed with normal diet, with fi'eedom diet and drinking, for 60 days, and then, the rats were pul to death so as to Lake out testis, the expression of transforming growth factor (TGF-β1) and epidermal growth factor (EGF) of testicular tissue was tested by immunohistochemistry. Results (1)The expression of EGF of testieular tissue in model group was less than other treatment groups, with significant differences (P 〈 0.05). (2)The expression of TGF-β1 of testieu- lar tissue in model group was more than other treatment groups, with significant differences (P 〈 0.05 or P 〈 0.01). Conclusion The changes of hwal growth factor EGF and TGF-β1 of testicular tissue in rats with experimental varieoeele during ptJherty are one of the possible nlechanisms that intluence lesticular spernmtogenetie function and lead to the decrease of sperm quality. Shugan Tongluo Qiangjing Deeoetion can ohviously down-regulate the expression of TGF-β1 and raise the exprcssion of El;F, so as to imlmwe the quality of semen, which may he one of the mechanisms of the proteclion of Shugan Tongluo Qiangjing Decoction in leslieular spermlogenetie function of testis in rats with improved experimenlal varieoeele during puherly.
