| 摘 要： | 目的测定雷公藤甲素微透析体外回收率及其影响因素。方法建立微透析样品中雷公藤甲素高效液相色谱法（HPLC）分析方法。利用增量法、减量法对比雷公藤甲素体外回收率，并考察流速、浓度对体外回收率的影响。结果所建立的高效液相色谱法，方法灵敏可靠，在所需浓度范围内线性良好；在同一浓度下，体外回收率随着流速的增大而减小；在同一流速下，浓度的改变对体外回收率的影响不大。结论本研究确定了影响微透析体外回收率因素，反透析法可作为微透析研究雷公藤甲素体外回收率的测定方法。

| Abstract: | Objective To investigate in-vitro recovery rate and influencing factors of triptolide in microdialysis probe. Methods The method used for the determination of triptolide in microdialysate samples by HPLC was established. The in-vitro recoveries were obtained by two approaches including recovery estimation by gain and loss methods. The ef- fect of flow rates and concentrations on recovery was studied. Results The method of HPLC was sensitive and reliable. The linearity correlation was good. In-vitro recovery rate was reduced by exponent law as the flow rate increased. At the same flow rate, the concentration had no obvious effect on in-vitro recovery. Conclusion This study confirms the influencing factors of in-vitro recovery rate of microdialysis probe. Retrodialysis method can be used to detect in-vitro recovery rate of triptolide. |
