大鼠急性脊髓损伤后tau蛋白表达规律及意义

摘    要:目的:探讨急性脊髓损伤后Wistar大鼠脊髓组织中tau蛋白动态表达规律及意义.方法:健康Wistar大鼠56只,雌雄不限随机分为两组,其中1组用改良Allen法制备成急性脊髓损伤模型,即作为手术组(48只);另1组则作为对照组(对照组仅开椎板,不损伤大鼠脊髓组织n=8).分别在12h、1d、3d、5d、7d、10d取出大鼠在T10及部分T9、T11椎板下的损伤脊髓组织制作蜡块并切片.采用HE染色及免疫组化法测定大鼠脊髓损伤后受损脊髓组织中tau蛋白的表达.结果:Wistar大鼠在急性脊髓损伤后,受损脊髓组织内即出现tau的阳性表达,随着时间的延长,tau蛋白的阳性表达结果逐渐增多;伤后3d阳性表达到达最大表达值,10d表达仍呈阳性且高于对照组.结论:损伤大鼠的脊髓组织后,组织中tau蛋白表达明显增加,且在不同时间点上有一定的变化关系,为急性脊髓损伤后临床的早期干预时机提供了一定的理论依据.

The pattern and significance of tau protein expression of Wistar rats after spinal cord injury

Abstract:Objective. To investigate dynamic expression of tau protein and its significance of spinal cord tis- sue of Wistar rats after acute spinal cord injury. Method. 56 Wistar rats of either sex were randomly divided in- to two groups, one group was prepared using a modified Allen into acute spinal cord injury model, that is, as surgery group (48); the other group was served as a control group (control group, only to open the lamina, does not damage the spinal cord tissue n~8). We removed injured spinal cord tissue under T10 and some Tg, Tll laminectomy in 12h, ld, 3d, 5d, 7d, 10d respectively and produced paraffin blocks and slices. HE staining and Immunohistochemical methods were used to determinate the expression of tau protein in injured spinal cord tissue. Results. Tau positive expression was appeared in injured spinal cord tissue after acute spinal cord injury on Wistar rats. The positive expression of Tau protein increased gradually as time went by and reached the maximum value after 3d injury. The expression continued to show positive after 10d and still higher than the control group. Conclusion. The tau protein expression was significantly increased in spinal cord tissue after it was damaged, and there are some changes on the relationship at different points in time, which provides a theo- retical basis for early intervention after acute spinal cord injury.
