红花注射液预处理对局灶性脑缺血再灌注大鼠脑组织中超氧化物歧化酶活性及丙二醛和兴奋性氨基酸含量的影响

摘    要:目的探讨红花注射液预处理对大鼠局灶性脑缺血再灌注损伤的保护作用。方法采用线栓塞法阻断大鼠右侧大脑中动脉,制备大鼠局灶性脑缺血再灌注损伤模型。40只大鼠随机分为对照组、假手术组、模型组和红花预处理组,每组10只。检测各组大鼠脑组织中超氧化物歧化酶(SOD)活性及丙二醛(MDA)、谷氨酸(Glu)、门冬氨酸(Asp)含量。结果对照组和假手术组大鼠脑组织中SOD活性及MDA、Glu、Asp含量比较差异均无统计学意义(P>0.05);与对照组和假手术组比较,模型组大鼠脑组织中SOD活性显著降低(P<0.05),MDA、Glu、Asp含量显著升高(P<0.05);与模型组比较,红花预处理组大鼠脑组织中SOD活性显著升高(P<0.05),MDA、Glu、Asp含量显著降低(P<0.05)。结论红花注射液对大鼠局灶性脑缺血再灌注损伤具有一定的保护作用,其机制可能与抑制脑组织脂质过氧化反应及兴奋性氨基酸有关。

Abstract:Objective To study the protection of safflower injection pretreatment on the rats with focal cerebral ischemical reperfusion injury. Methods The models of rats with focal cerebral ischemical reperfusion injury were made by blocking the right middle cerebral artery occlusion.Forty rats were randomly divided into control group,sham operation group,model group and safflower pretreatment group(safflower group),ten rats in each group.The activity of superoxide dismutase(SOD) and the contents of malondialdehyde(MDA),glutamic acid(Glu) and aspartic acid(Asp) in all rats were detected. Results There was no statistical difference in the SOD activity and MDA,Glu,Asp contents of brain tissue between control group and sham operation group(P>0.05).Compared with control group and sham operation group,the activity of SOD was significantly decreased(P<0.05),moreover,the contents of MDA,Glu and Asp were significantly increased in brain tissue of rats in model group(P<0.05).Compared with model group,the activity of SOD was significantly increased(P<0.05),moreover,the contents of MDA,Glu and Asp were significantly decreased in brain tissue of rats in safflower group(P<0.05). Conclusion Safflower injection on focal ischemical reperfusion injury rats has a protective effect,the mechanism may be related to inhibition of brain tissue lipid peroxidation and excitatory amino acids.
