盐酸纳络酮对急性酒精中毒小鼠肝组织的影响

摘    要:目的探讨盐酸纳络酮对急性酒精中毒小鼠血清谷丙转氨酶(ALT)和肝脏组织的影响。方法30只小鼠随机分为对照组、模型组和盐酸纳络酮组,每组10只。对照组用蒸馏水灌胃,模型组和盐酸纳络酮组均用酒精灌胃,以翻正反射消失为急性酒精中毒标准,对盐酸纳络酮组急性酒精中毒小鼠注射盐酸纳络酮注射液(0.4 g.L-1)0.3 mL。处理结束后,摘出眼球取血,取肝组织。测血清ALT活力,制作肝组织病理切片,显微镜下观察肝脏细胞受损情况。结果模型组和盐酸纳络酮组小鼠血清ALT活力高于对照组(P<0.05,P<0.01),盐酸纳络酮组ALT活力高于模型组(P<0.05)。病理切片显示对照组小鼠肝细胞结构完整且均匀,模型组小鼠肝细胞有不同程度损伤,盐酸纳络酮组小鼠肝细胞损伤最严重。结论盐酸纳络酮注射液不能逆转高剂量酒精对肝组织的损伤,相反会加重肝细胞代谢压力。

Abstract:Objective To explore the effect of naloxone hydrochloride on alanine aminotransferase(ALT) in serum and hepatic tissue in acute alcoholism mice.Methods Thirty mice were divided into control group(n=10),model group(n=10) and naloxone hydrochloride group(n=10).Water was lavaged in the control group,while alcohol were lavaged in the model and the naloxone hydrochloride group.With the disappear of righting reflex as the standard of acute alcoholism,naloxone hydrochloride injection(0.4 g·L-1,0.3 mL) was injected in the naloxone hydrochloride treating group.After treatment,the eyeballs were removed,and the blood and hepatic tissue was collected.The activity of ALT was detected,the pathology slices of liver tissue were made,and the changes of damaged hepatic tissue were observed by microscope.Results The activity of ALT in the model group and the naloxone hydrochloride group were significantly higher than that in the control group(P<0.05,P<0.01),and the activity of ALT in the naloxone hydrochloride group was higher than that in the model group(P<0.05).The pathology slices showed that the structural of hepatic cell were complete and uniform in the control group,the hepatic cell were damaged to some extent in the model group,and the hepatic cell were most seriously damaged in the naloxone hydrochloride group.Conclusion Naloxone hydrochloride cannot alleviate the damage of hepatic tissue induced by alcohol of high concentration,on the contrary,it aggravates the damage of hepatic tissue.
