鼠脑损伤早期脑微血管多巴胺含量的变化及其临床意义

摘    要:目的探讨大鼠脑损伤早期脑微血管多巴胺的变化及其临床意义。方法用改良的Fe-ney’s法制作脑损伤模型,应用干-湿重法检测脑组织水含量,利用高效液相色谱-电化学检测器检测大鼠脑损伤不同时程脑微血管(brainmicrovesels,BMVs)中多巴胺(dopamine,DA)的含量。结果脑损伤后0.5h,脑微血管中DA含量与对照值相比显著增加(P<0.01),24h仍显著高于对照值(P<0.01);脑损伤后0.5h脑皮质及脑白质水含量显著高于对照值(P<0.01),伤后24h达高峰,与对照值相比差别显著(P<0.01)。结论脑微血管DA与创伤性脑水肿的发生、发展有密切关系。

Abstract:Objective To research the changes of dopamine contents in BMVs and their influence on cerebral edema at the early stage of brain injury.Methods Brain injury model was made through an improved Feeney's method.The dopamine contents in the BMVs were measured by using high performance liquid chromatogrphy with electrochemical detector(HPLC-FCD),and the water contents of brain tissue were measured by dry-wet method.Results The contents of DA in the BMVs increased significantly at 0.5h,and were still higher than the control values at 24h after brain injury( P <0.01);the water content of grey and white matters increased remarkedly at 0.5h( P <0.01);the augmentaion of water content of the grey and white matters was most remarkably at 24h after brain injury ( P <0.01).Conclusion Dopamine might be one of the most important fractors in the production and development of traumatic brain edema.
