盐酸法舒地尔在脂肪干细胞向表皮细胞诱导分化中的作用

摘    要:目的探讨盐酸法舒地尔(HA1077)在脂肪干细胞(ADSC)向表皮细胞诱导分化中的作用。方法用酶消化法消化人脂肪组织获取ADSC,以基础培养基即体积分数含10%胎牛血清的DMEM培养基传代,取生长良好达到90%融合的第3代人ADSC分为4组,第1组持续用基础培养基培养、传代;将第2、3、4组弃基础培养基,无菌磷酸盐缓冲溶液冲洗,分别换用培养液2(基础培养基+20μmol.L-1 HA1077)、培养液3(体积分数70%的基础培养基+体积分数30%的成纤维细胞培养基上清液+10 ng.L-1重组人表皮生长因子)、培养液4(20μmol.L-1 HA1077+培养液3),诱导后经免疫细胞化学、流式细胞仪检测CK19的表达。结果免疫细胞化学分析表明ADSC表达CD44、CD49d,而不表达CD106、CD34、CK19;流式细胞仪检测CD44、CD49d的阳性率分别为(98.84±0.02)%、(92.52±0.04)%。流式细胞仪检测各组CK19的表达阳性率分别为(0.23±0.01)%、(0.35±0.02)%、(9.73±0.80)%、(17.65±1.00)%,其中第3、4组CK19的阳性率较对照组明显增高,差别均有统计学意义(P<0.05)。结论 HA1077对ADSC向表皮细胞诱导分化有促进作用。
