川芎嗪对大鼠肾缺血再灌注损伤的保护作用

摘    要:目的探讨川芎嗪(LT)对大鼠急性肾缺血再灌注损伤的保护作用及其机制。方法28只SD大鼠随机分为4组,(每组7只),分别为正常对照组、假手术组、缺血再灌注组、治疗组。采用双侧肾蒂夹闭的肾缺血再灌注损伤模型,测定肾组织丙二醛(MDA)水平和超氧化物歧化酶(SOD)的活性,同时测定血清尿素氮(BUN)和肌酐(Cr)水平,并在光镜下观察肾组织病理变化。结果肾缺血再灌注组与对照组和假手术组相比,MDA、BUN和Cr含量明显升高(P<0.01),SOD活性下降(P<0.05),治疗组MDA、BUN和Cr含量下降(P<0.05),SOD活性增强(P<0.05)。结论川芎嗪对急性肾缺血再灌注损伤具有保护作用,其机制可能与抗自由基损伤有关。

Abstract:Objective To investigate the protection and mechanism of ligustrazine(LT) on renal ischemic reperfusioninjury in rats. Methods 28 rats were randomly divided into 4 groups.(7 rats were used in each group),including of control group ,sham operation group,ischemic reperfusion group and treatment group. Bilateral renal ischemia was induced by occluding the renal pedicles for 45 min following reperfusion. Malondialdehyde(MDA) levels and superoxide dismutase(SOD) activities were determinded to assess lipid peroxidation. BUN and Cr were also determinded to assess the function of the kidney. Pathologic changes were studied by light microscopy. Results Compared with the control and sham groups, the contents of MDA,BUN and Cr rose dramatically in the ischemia reperfusion group(P<0.01), while the activity of SOD decreased(P<0.05). But in the treated group, the contents of MDA,BUN and Cr decreased(P<0.05), while the activation of SOD rose(P<0.05). Conclusion LT may protect the kidney of acute ischemia and reperfusion injury in rats. The mechanism may be related to anti-lipid peroxidation.
