摘 要: | 目的观察黄芪多糖对高脂饮食诱导的大鼠胰岛素敏感性和脂肪细胞因子的影响。方法将健康雄性SD大鼠随机分为正常对照组(n=12)和高脂模型组(n=36),分别给予普通饲料和高脂饲料。模型建立成功后,高脂模型组随机分为高脂对照组、盐酸吡格列酮组和黄芪多糖组,每组12只。分别给予生理盐水、盐酸吡格列酮20mg.kg-1.d-1和黄芪多糖400mg.kg-1.d-1,干预8周。随机选取各组中的一半大鼠进行高胰岛素-正葡萄糖钳夹实验,测定稳态期葡萄糖输注率(GIR)。同时测定另一半大鼠空腹胰岛素(FINS)、血脂、白细胞介素6(IL-6)、瘦素、抵抗素、超敏C反应蛋白(hs-CRP)、脂联素(APN)、肿瘤坏死因子-α(TNF-α)及游离脂肪酸(FFA)水平。结果与正常对照组比较,高脂对照组GIR降低(P<0.01);与高脂对照组比较,盐酸吡格列酮组和黄芪多糖组GIR升高,差异均有统计学意义(P<0.01)。与正常对照组比较,高脂对照组FINS、FFA、总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白(LDL)、IL-6、瘦素、抵抗素、hs-CRP及TNF-α升高,高密度脂蛋白(HDL)、APN降低,差异均有统计学意义(P<0.01)。与高脂对照组比较,盐酸吡格列酮组和黄芪多糖组的FINS、FFA、TC、TG、LDL、IL-6、瘦素、抵抗素、TNF-α降低,APN升高,差异有统计学意义(P<0.05,P<0.01)。结论黄芪多糖可以提高大鼠胰岛素敏感性,改善其胰岛素抵抗,其机制可能与其调节血脂水平和脂肪细胞因子水平有关。

Effect of astragalus polysaccharides on insulin sensitivity and adipocytokines in rats |

Abstract: | Objective To observe the effect of astragalus polysaccharides(APS) on insulin sensitivity and adipocytokines in rats.Methods Forty-eight male SD rats were randomly divided into normal control group(n=12) and high fat-diet-induced model group(n=36).Rats in normal control group were fed with ordinary diet while those in high fat-diet-induced model group were fed with high fat diet.Then the rats in high fat-diet-induced model group were randomly divided into high fat-diet-induced control group,pioglitazone group and APS group,with 12 rats in each group;normal saline,pioglitazone(20 mg·kg-1·d-1) and APS(400 mg·kg-1·d-1)were lavaged respectively for eight weeks.Changes of fasting insulin(FINS),plasma lipid levels,interleukin-6(IL-6),leptin,resistin,hypersensitive C-reactive protein(hs-CRP),adiponectin(APN),tumor necrosis factor-α(TNF-α) and free fatty acids(FFA) in one half rats of each group were routinely measured,meanwhile glucose infusion rate(GIR) of another half rats was evaluated by hyperinsulinaemic-euglycaemic clamp technique.Results Compared with the normal control group,the GIR in high fat-diet-induced control group was lower(P<0.01).Compared with high fat-diet-induced control group,the GIR in pioglitazone group and APS group were higher,the differences were statistical significance(P<0.01).Compared with normal control group,the FINS,FFA,total cholesterol(TC),triglyeride(TG),low-density lipoprotein(LDL),IL-6,leptin,resistin,hs-CRP and TNF-α of the rats in high fat-diet-induced control group were significantly higher(P<0.05),while high density lipoprotein(HDL) and APN were significantly lower(P<0.05).Compared with high fat-diet-induced control group,the FINS,FFA,TC,TG,LDL,IL-6,leptin,resistin and TNF-α of the rats in pioglitazone group and APS group were lower,while APN was higher(P<0.05,P<0.01).Conclusion APS can increase the insulin sensitivity of rats and improve its insulin resistance,which mechanism may be in relation to adjust blood lipid level and adipose cell factor level. |
