白藜芦醇对骨关节炎模型兔关节液中一氧化氮水平的影响

目的探讨白藜芦醇对骨关节炎(OA)模型兔关节液中一氧化氮(NO)水平的影响。方法36只健康新西兰兔随机分为正常组、模型组、阳性组和白藜芦醇高、中、低剂量组。模型组、阳性组和白藜芦醇组用Hu lth法复制实验性骨关节炎模型。术后8周造模成功,阳性组用硫酸氨基葡萄糖35 mg.kg-1灌胃,白藜芦醇高、中、低剂量组分别给予白藜芦醇120 mg.kg-1、60 mg.kg-1、30 mg.kg-1灌胃,灌胃6周后处死兔并切取股骨内髁软骨作光镜观察,测定骨关节液中NO含量并作统计学处理。结果模型组兔关节软骨呈明显退行性变,模型组关节液NO含量显著高于白藜芦醇组(P<0.05),灌胃后阳性组和白藜芦醇组关节液NO含量无显著性差异(P>0.05)。结论白藜芦醇能防止并延缓关节软骨的退行性改变。

Effect of Resveratrol on nitric oxide in the synovial fluid in rabbits with osteoarthritis

Objective To explore the effects of Resveratrol on nitric oxide(NO) in the synovial fluid in rabbits with osteoarthritis(OA).Methods Thirty-six New Zealand rabbits were divided randomly into the normal group,model group,positive control group and Resveratrol high,middle,low dosage group.The model group,positive control group(35 mg·kg~(-1))and Resveratrol high(120 mg·kg~(-1)),middle(60 mg·kg~(-1)),low(30 mg·kg~(-1))dosage group established the OA model with Hulth′s method.Eight weeks later,the model were successfully established,and Resveratol perlavage was given drugs for 6 weeks.Then all animals were sacrificed.The articular cartilage of medial femoral condyle was observed at macroscopically and light microscopically.The content of NO in the synovial fluid was measured by nitrate reduction method.Results Articular cartilage degeneration was obviously in model group.The level of NO in the synovial fluid in Resveratrol group was significantly lower than that in the model group(P<0.05).Conclusion Resveratrol can prevent and alleviate the degradation of articular cartilage.
