摘    要:目的探讨链脲菌素(STZ)实验性糖尿病大鼠组织钙调素(CaM)活性的变化及其对胰岛素治疗的反应。方法Wistar大鼠32只,分为正常对照组、糖尿病非治疗72h病程组、2wk病程组及胰岛素治疗组。用STZ制成糖尿病模型。采用依赖于Ca2+的磷酸二酯酶分析法测定肝脏、脂肪、骨骼肌和肾脏组织中可溶性钙调素活性。结果①与正常对照组相比,各糖尿病非治疗组组织中CaM活性均显著降低,平均下降40%~60%不等,不同病程组间无显著差异。胰岛素治疗可使肝脏、脂肪和骨骼肌组织中的CaM活性恢复正常。②各组织中活性与血清胰岛素水平均呈正相关。结论Ca2+-CaM系统在胰岛素作用机制和糖尿病的发生机理中,可能起重要作用。

Abstract:Objective To research the changes of the calmodulin (CaM) activity and its response to the insulin treatment in streptozotocin-induced diabetic rat tissues.Methods Thirty two wistar rats were divided into normal control group, 72-hour untreated diabetic group, two-week untreated diabetic group and insulin-treated diabetic group. The diabetes model was established in rats by peritoneal injection of streptozotocin. The CaM activity of the tissues (liver, fat, skeletal muscle and kidney) was measured by Ca2+-dependent phosphodiesterase assay.Results ①Compared it with the normal control group, the CaM activiy of all the tissues in untreated diabetic groups was reduced obviously (decreased 40%-60% ), which could be restored foward normal after insulin administration, especially in liver, adipose and skeletal muscle tissues. There was no difference between the two untreateel groups. ②The CaM activity of all the tissues has a positive correlation within sulin level in serum.Conclusion Ca2+-CaM system may play an important role in the mechnism of insulin action and in the pathogenesis of diabetes mellitus.
