邻苯二甲酸二乙己酯及双酚A联合染毒对大鼠肝脏的影响

摘    要:目的研究邻苯二甲酸二乙基己酯(Di-2-ethylhexyl phthalate,DEHP)和双酚A(Bisphenol A,BPA)联合暴露对大鼠肝脏的影响。方法将Wistar大鼠随机分4组,分别灌胃1 500mg/kg DEHP、200mg/kg BPA及750mg/kg DEHP+100mg/kg BPA染毒6周。染毒终点测定血清中碱性磷酸酶(ALP)、胆固醇(CHO)、甘油三酯(TG)及镁离子(Mg2+)含量;检测肝匀浆中谷胱甘肽(GSH)、丙二醛(MDA)含量。结果与对照组比较,DEHP明显升高血清ALP、CHO及Mg2+含量,而BPA明显增加肝中MDA含量(均P〈0.05)。与对照组比较,DEHP+BPA联合染毒除明显增加肝脏脏器系数外,对大鼠体重、肝MDA、血脂及血清Mg2+和ALP含量均无明显影响。与DEHP+BPA联合染毒组比较,BPA显著升高肝脏MDA含量;DEHP增加血清Mg2+含量差异均有统计学意义(均P〈0.05)。结论 DEHP和BPA单独染毒均可产生肝损伤,其联合暴露产生交互作用。

COMBINED EFFECT OF PHTHALATE AND BPA ON LIVER IN RATS

Abstract:Objective To study the combined effect of diethylhexyl phthalate(DEHP)and bisphenol A(BPA)on liver in rats.Methods All the Wistar rats were randomly divided into 4 groups and respectively were gavaged for 6-week with1 500mg/kg DEHP,200mg/kg BPA and 750mg/kg DEHP plus 100mg/kg BPA.The concentrations of alkaline phosphatase(ALP),cholesterol(CHO),triglycerides(TG)and magnesium(Mg2+)in serum were determined.The contents of glutathione(GSH)and malondialdehyde(MDA)in liver homogenate were detected.Results Compared with the control group, DEHP significantly increased the content of ALP,CHO and Mg2+in serum,and the concentration of hepatic MDA was significantly higher than the BPA group(all P〈0.05).DEHP+BPA had no obviously effect on rat weight,MDA concentration in liver homogenate,lipids,Mg2+and ALP in serum other than the coefficient of liver.Compared with DEHP+BPA combined exposure group,there was significant increase in MDA concentration in liver homogenate of BPA group and Mg2+of DEHP group(P〈0.05).Conclusion Both DEHP and BPA can induce liver damage,and the combined exposure has interaction effect.
