抵抗素基因单核苷酸多态性与2型糖尿病的相关性研究

摘    要:目的探讨抵抗素基因rs34861192单核苷酸多态性与2型糖尿病的关系。方法分别测定2型糖尿病组(T2DM组)100例及正常对照组(CON组)150例空腹血糖(FBG)、空腹胰岛素(FIN)、甘油三酯(TG)、总胆固醇(CHOL)、高密度脂蛋白胆固醇(HDL-CH)、低密度脂蛋白胆固醇(LDL-CH)和抵抗素,并计算胰岛素抵抗指数(IR-I);同时采用基质辅助激光解吸电离飞行时间质谱(MALDI-TOFMS)技术对T2DM组和CON组进行抵抗素rs34861192位点基因型的检测,根据结果进行综合分析。结果T2DM患者FBG、FIN、IRI、TG、抵抗素的检测数值分别为(9.96±5.86)mmol/mL,(12.23±9.21)μg/mL,6.25±4.13,(2.02±1.85)mmol/mL,(7.24±3.65)ng/mL]高于健康人群的检测数值分别为(4.94±0.50)mmol/mL,(5.6±2.66)μg/mL,1.24±0.62,(1.33±0.74)mmol/mL,(1.44±0.93)ng/mL],差异有统计学意义(t值分别为8.535,4.586,4.936,3.516,15.524,P均〈0.05);T2DM组和CON组抵抗素rs34861192位点的基因型分布分别为AA6%、AG40%、GG54%和AA0.7%、AG20%、GG79.3%;抵抗素基因rs34861192位点的基因型分布和等位基因频率在T2DYl组和CON组问有统计学意义(x。值分别为20.231、19.103,P均〈0.05)。结论抵抗素可能是T2DM的危险因子,抵抗素基因rs34861192位点多态性与2型糖尿病相关,同时可能对脂代谢有一定影响。

Abstract:Objective To research the relationship between the single nucleotide polymorphism of resistance gene rs34861192 with incidence of type 2 diabetes.Methods Respectively measured fasting plasma glucose(FBG),fasting in-sulin(FIN),triglyceride(TG),total cholesterol(CHOL),high density lipoprotein cholesterol(HDL-CH),low density lipoprotein cholesterol(LDL-CH) and resistin levels in T2DM group(100 cases) and the CON group(150 cases),then calculated the insulin resistance index(IRI).While used matrix secondary laser solutions sucking ionization flight time mass spectrum(MALDI-TOF MS) technology to test the genotype of resistance gene rs34861192 in T2DM group and CON group.Results FBG,FIN,IRI,TG and resistin levels (9.96±5.86) mmol/mL,(12.23±9.21) μg/mL,6.25±4.13,(2.02± 1.85) mmol/mL and(7.24±3.65) ng/mL) ] in T2DM group were significantly higher than CON group (4.94±0.50) mmol/mL,(5.63±2.66) μg/mL,1.24±0.62,(1.33±0.74) mmol/mL and(1.44±0.93)n g/mL].The differences were statistically significant(t=8.535,4.586,4.936,3.516 and 15.524,all P < 0.05);The resistance gene rs34861192 genotype distribution in T2DM group and CON group respectively,were AA6%、AG40%、GG54% and AA 0.7%、 AG20%、GG79.3%.The genotype distri-bution and the allele frequency of resistance gene rs34861192 were significant different between the T2DM group and CON group(χ2= 20.231 and 19.103,both P < 0.05).Conclusion Resistin may be a risk factor in type 2 diabetes;Resistance gene rs34861192 polymorphism is related with the incidence of type 2 diabetes,and may have some influence on lipid metabolism.
