乌司他丁和血必净抑制早期内毒素血症大鼠炎症反应的对比研究

摘    要:目的研究乌司他丁(ulinastatin,UTI)抑制早期内毒素(LPS)血症大鼠的炎症反应是否优于血必净(Xue-bijing,XBJ)。方法通过鼠尾静脉注射LPS复制LPS血症大鼠模型。54只SD大鼠随机分为LPS组、(LPS+UTI)组、(LPS+XBJ)组,每组18只,分别在用药后8 h、24 h、48 h时点随机快速处死6只大鼠,取心脏血分离血清,用双抗体夹心酶联免疫吸附法测定白细胞介素-1(IL-1)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)。结果三组中各时间点TNF-α、IL-6、IL-1分别比较差异均有统计学意义(P<0.05);(LPS+UTI)组在8 h时间点与(LPS+XBJ)组比较差异有统计学意义(P<0.05),在24 h、48 h时间点差异无统计学意义(P>0.05)。结论 UTI和XBJ均能有效抑制LPS大鼠的炎症反应;UTI在炎症早期抑制炎症反应优于XBJ。

Comparative study on Ulinastatin and Xuebijing to inhibit early inflammatory response in endotoxemia rats

Abstract:Objective To investigate whether Ulinastatin (UTI) is better than Xuebijing(XBJ) in inhibiting inflamma tory response of endotoxemia(LPS) rats. Methods Rats were rendered LPS model when injected intravenously with LPS. 54 SD rats were randomly divided into three groups: LPS group, (LPS+UTI) group and (LPS+XBJ) group. Each group consisted of 18 rats. Six rats in each group were killed randomly and the heart blood was taken out at 8 h, 24 h and 48 h respectively, and then serums were separated, and a double-antibody sandwich enzyme-linked immunosor- bebt assay was applied to detect tumor necrosis factor-or (TNF-α), Intedeukin-6 (IL-6) and Intedeukin- 1 (IL- 1 ). Results The differences among the TNF-α,IL-6 and IL-1 of LPS group, (LPS+UTI) group and (LPS+XBJ) group at all time points were of statistical significance (P 〈 0.05) ;At 8 h the differences between (LPS+UTI)group and (LPS+ XBJ) group were statistically significant (P 〈 0.05); But those were not statistically significant at 24 h and 48 h (P 〉 0.05). Conclusion Both UTI and XBJ can inhibit the inflammatory response in LPS rats effectively; but UTI is bet- ter than XBJ in inhibiting the early inflammatory response.
