电项针对失眠大鼠脑干单胺类神经递质含量的影响

摘    要:目的 通过观察不同刺激频率电项针对PCPA致失眠大鼠脑干内单胺类神经递质5-羟色胺(5-HT)、多巴胺(DA)和去甲肾上腺素(NE)含量的影响,初步探讨电项针促眠的作用机制及不同刺激频率电项针的疗效差异.方法 将Wistar大鼠50只随机分成5组,即正常组,模型组,电项针1组(取刺激频率2 Hz)、电项针2组(取刺激频率50Hz)、电项针3组(取刺激频率100Hz).除正常组外,各组采用腹腔注射对氯苯丙氨酸(PCPA)制备大鼠失眠模型.造模成功后,电项针各组分别按各自刺激频率给予干预治疗,1次/d,30 min/次,连续7 d,治疗后留取脑干组织检测单胺类神经递质5-HT、DA、NE的含量变化.结果 模型组大鼠脑干组织中5-HT含量明显低于正常组,DA、NE含量高于正常组,差异具有统计学意义(P<0.05);经电项针治疗7 d后,失眠大鼠脑干5-HT含量升高,DA、NE含量显著降低,与模型组比较差异有统计学意义(P<0.05);电项针各组之间疗效比较,电项针1组明显优于电项针2和电项针3组(P<0.05);电项针2和电项针3组比较无统计学意义(P>0.05).结论 电项针改善失眠的作用机制可能与调节失眠大鼠脑干内单胺类神经递质5-HT、DA、NE的质量浓度有关;电项针2组疗效优于其他2组,说明2Hz刺激频率电项针治疗失眠效果更好.

Research of electro-nape-acupuncture on the contents of monoamine neurotransmitters in brainstem of insomnia rats

Abstract:Objective To observe the effect of different frequencies of electro-nape-acupuncture (ENA) on the contents of monoamine neurotransmitters 5-hydroxytryptamine (5-HT), dopamin (DA) and norepinephrine (NE) in brainstem of insomnia rats induced by PCPA, to discuss primarily the mechanism of promoting sleep and therapeutic efficacy of different frequencies of ENA. Methods Fifty Wistar rats were randomly divided into five groups, those are normal group, model group, ENA group Ⅰ (stimulus frequency of 2 Hz), ENA group Ⅱ (stimulus frequency of 50 Hz) and ENA group Ⅲ (stimulus frequency of 100 Hz). The animal models of insomnia were established by para-chlorophenylalanine (PCPA) intraperitoneal injection in each group except normal group. After duplicating the models successfully, each ENA group rats were treated by ENA therapy at different stimulus frequencies respectively for seven consecutive days, 1 time/day, 30min/time. After treatment the brainstems were taken and measured the contents of neurotransmitters 5-HT, DA and NE. Results The content of 5-HT were significantly lower and the contents of DA and NE were higher in the animal model of rats than that in the normal group, there were statistically significant differences (P<0.05); After the seven days of ENA treatment, the 5-HT content increased and the DA and NE contents decreased in brainstems of insomnia rats. Compared with the model group, there were significant differences (P<0.05). Among the ENA groups, ENA group Ⅰ was greatly superior to ENA group Ⅱ and ENA group Ⅲ at regulating the levels of monoamine neurotransmitters (P<0.05), however, there were no statistically significant difference between ENA group Ⅱ and ENA group Ⅲ (P >0.05). Conclusion The mechanism of ENA improve the structure of sleep may involve the regulation of monoamine neurotransmitters 5-HT, DA and NE contents in brainstem of insomnia rats.
