桂枝甘草汤对运动性贫血大鼠血浆总超氧化物歧化酶和血清铁的影响

摘    要:目的探讨桂枝甘草汤对运动性贫血大鼠血浆总超氧化物歧化酶(SOD)活性和血清铁浓度的影响。方法取25只SPF级SD雄性大鼠随机分为空白对照组(Control组)、乙酰苯肼组(APH组)、运动性贫血组(APH+S组)、桂枝甘草汤1组(APH+S+GGD 1组)、2组(APH+S+GGD 2组)。除Control组注射生理盐水外,其余各组大鼠均注射2%乙酰苯肼(APH),其中APH+S、APH+S+GGD 1组和APH+S+GGD 2组大鼠进行每日力竭游泳运动,同时这3组大鼠分别灌胃生理盐水、0.018 g/m L和0.036 g/mL的桂枝甘草汤水煎剂。在给药后第15天取眼后静脉丛血进行全血分析检测;利用血浆总SOD和血清铁试剂盒进行血浆总SOD活性与血清铁浓度检测。结果全血分析结果显示,APH+S+GGD 1组和APH+S+GGD 2组大鼠红细胞计数(RBC)明显高于APH、APH+S组(P0.05);APH+S+GGD 2组大鼠红细胞平均体积(MCV)明显低于APH和APH+S组(P0.05);血浆总SOD活性与测试结果中的液体超氧化物歧化酶(SOD)活性呈负相关,从液体中SOD活性可得出血浆总SOD活性结果为:APH+S+GGD 1组和APH+S+GGD 2组大鼠血浆总SOD活性明显低于APH+S组(P0.05);血清铁检测结果发现,APH+S+GGD 1组大鼠血清铁浓度明显高于APH组(P0.05);APH+S+GGD 1组和APH+S+GGD 2组大鼠血清铁浓度明显高于APH+S组(P0.05)。结论桂枝甘草汤可以明显改善运动性贫血所致的红细胞减少;同时,可明显缓解运动性贫血大鼠的血浆总SOD活性代偿性增高以及降低运动性贫血大鼠血清铁浓度,说明桂枝甘草汤可以通过改善血红蛋白携氧能力以及抗氧化保护红细胞来改善运动性贫血大鼠的健康状态。同时,为桂枝甘草汤的临床应用提供实验依据。

Abstract:Objective To investigate the effect of Guizhi Gancao decoction on plasma superoxide dismutase (SOD) activity and serum iron concentration in rats with exercise anemia.Methods 25 SPF male SD rats were randomly divided into control group, the acetylphenylhydrazin (APH) group, the exercise anemia group(group APH+S), the Guizhi Gancao decoction 1 group (group APH+S+GGD 1)Guizhi Gancao decoction 2 group (APH+S+GGD 2).Except the control group whose rats were injected physiological saline, the remaining rats were injected 2 g/mL APH. APH+S, APH+S+GGD 1, APH+S+GGD 2 group rats were forced to do daily exhaustive swimming exercise, they were respectively intragastric administration with physiological saline, concentration of 0.018 g/mL and 0.036 g/mL Guizhi Gaocao decoction simultaneously. At the fi fteenth day after administration, the blood samples collected from the retinal vein were used as whole blood analysis. After that, the plasma from abdominal aorta were measured by the total SOD activity detection kit and serum iron detection kit.Results The whole blood analysis shown that the red blood cell count (RBC) in APH+S+GGD 1 and APH+S+GGD 2 group was signifi cantly higher than that of APH and APH+S group (P<0.05); The mean corpuscular volume (MCV) of the APH+S+GGD 2 group was significantly lower than that of APH and APH+S group (P<0.05); The plasma total sod activity was negatively correlated with the liquid superoxide dismutase (SOD) activity. From the sod activity, the plasma total sod activity was obtained as follows: The total SOD activity in APH+S+GGD 1 and APH+S+GGD 2 groups was significantly lower than that in APH+S group (P<0.05). The results of serum iron detection showed that the serum iron concentration in APH+S+GGD 1 group was significantly higher than that in APH group (P<0.05). The serum iron concentration in APH+S+GGD 1 and APH+S+GGD 2 group was significantly higher than that in APH+S group (P<0.05).Conclusion Guizhi Gancao decoction could obviously relieve erythropenia from exercise anemia; meanwhile, it could significantly decrease the activity of total plasma SOD and decrease the serum iron concentration in rats with exercise anemia, which states that Guizhi Gancao decoction could improve health condition of exercise anemia rats by increasing oxygen carrying capacity and decreasing compensation of oxidation resistance, and provides experimental evidence for clinical application of Guizhi Gancao decoction.
