电针对2型糖尿病大鼠右心房自主神经损害的调节作用

摘    要:目的 探讨针刺对2型糖尿病大鼠右心房自主神经损害的调节水平,为针刺临床优化选穴提供参考依据.方法 60只SD雄性大鼠随机分为4组,空白对照组、DM模型组、电针1组、电针2组.除空白对照组外,其余3组给予高脂高糖喂养4周后,用STZ 25 mg/kg连续2 d腹腔注射制作实验性糖尿病大鼠模型.造模2周后,空白对照组和DM模型组不予治疗;电针1组:选内关、曲池、后三里、三阴交;电针2组:选肺俞、心俞、胰俞、脾俞、肾俞,1次/d,6 d为1个疗程,连续治疗4个疗程,疗程间休息1 d.Elisa法检测4组大鼠右心房心肌中酪氨酸羟化酶(TH)、胆碱乙酰转移酶(ChAT)、神经生长因子(NGF)、生长相关蛋白-43(GAP-43)、睫状神经生长因子(CNTF)等含量.结果 与空白对照组比较,DM模型组ChAT显著下降(P<0.05),NGF有明显上升(P<0.05),GAP-43和CNTF有明显下降(P<0.05);与DM模型组相比较,2个电针组的ChAT有明显上升(P<0.05),NGF有明显下降(P<0.05),GAP-43和CNTF均有明显上升(P<0.05),且电针1组优于电针2组.结论 电针肢体腧穴对DM模型大鼠FPG有良好的调整作用,电针能够良性调节右心房TH、ChAT、NGF、GAP-43、CNTF的水平,减轻心脏自主神经纤维损害程度,且肢体腧穴组优于背俞穴组.

Regulating effect of electroacupuncture in the right atrium autonomic nerve damage of type 2 diabetic rats
Abstract:Objective To investigate the regulating effect of electroacupuncture on the autonomic neurotrophic factor in the right atrium of type 2 diabetic rats, and to provide reference for electroacupuncture clinical optimization. Methods Sixty SD rats were randomly divided into four groups: blank control group, DM model group, electroacupuncture group 1 and electroacupuncture group 2. Except the blank control group, the other three groups were fed with high fat and high glucose for 4 weeks. The experimental diabetic rats were injected intraperitoneally with STZ for 2 days at 25 mg.kg-1. After 2 weeks of modeling, the blank control group and DM model group were not treated; electroacupuncture 1 group: select Neiguan (PC6), Quchi (LI11), Zusanli (ST36), Sanyinjiao (SP6);electroacupuncture 2 group: select Feishu (BL13), Xinshu (BL15), Yishu (EX-B3), Pishu (BL20), Shenshu(BL23). Once a day, six days for a course of treatment, continuous treatment for four courses, one-day rest between the courses. TH, ChAT, NGF, GAP-43, and ciliary, Nerve growth factor (CNTF) in the right atrium of rats were assessed with ELISA. Results ChAT of DM group decreased significantly (P <0.05), NGF was increased significantly (P <0.05), GAP-43 and CNTF were significantly decreased (P < 0.05) in the DM model group compared with the control group;the ChAT of the two electroacupuncture groups increased significantly (P <0.05) NGF decreased significantly (P <0.05), GAP-43 and CNTF were significantly increased (P < 0.05) compared with DM model, electroacupuncture group 1 was superior to electroacupuncture 2 group. Conclusion Electroacupuncturing on limbs has a good effect on FPG of DM model rats, electroacupuncture can regulate the right ventricular autonomic neurotrophic factor balance, reduce the degree of autonomic nerve fiber damage and group of limbs acupoints better than that of group of back transport points.
