人脐血干细胞对裸小鼠移植的实验研究

摘    要:目的 探讨脐血干细胞移植到小鼠体内的增殖、分化以及维持情况。方法 将人脐血单个核细胞注入经致死量照射后的免疫缺陷小鼠— BAL B/C nu 小鼠。观察小鼠的生存、造血重建和植入指标情况。结果 移植组小鼠生存、造血重建及植入指标表达明显优于对照组 (P<0 .0 5 ) ,人脐血中的 CD34 细胞可植入且定居于小鼠骨髓中 ,并可在其中增殖、分化。结论 人脐血干细胞移植入裸鼠显示脐血的造血干 /祖细胞在体内具有长期重建造血的能力

Abstract:Objective The goal of this study is to evaluate the repopulating property and hematopoietic potent of human umbilical cord blood cells when transplanted into BALB/C nu mice.Methods CD34 cells from human umbilical cord blood were transplanted into lethally irradiated immunodeficient BALB/C nu mice and some important indices such as survival rate,hematopoiesis reconstitution and engraftment sign were recorded and analyzed.Results In terms of survival rate,hematopoiesis reconstitution and engraftment sign,the data collected from transplanted mice were significantly higher than those from control group,indicating the successful engraftment within murine bone marrow and subsequently effective proliferation and differentiation of CD34 cells.Conclusion Umbilical cord blood can be transplanted to null mice of different species.Hematopoietic progenitor/stem cells in human umbilical cord blood can reconstitute and sustain hematopoietic function in vivo.
