乌头碱水解实验和热力学研究

摘    要:目的探索乌头碱的水解机理,解释附子长时间煎煮解毒的机理;方法采用Am1半经验量子化学方法,对乌头碱水解反应的热力学进行了研究;采用HPLC和液质联用方法,对乌头碱水解产物进行了分析.结果理论上预测了双酯型乌头碱较易生成苯甲酰乌头碱,实验上证实了双酯型乌头碱主要水解为单酯型苯甲酰乌头碱.结论单酯型苯甲酰乌头碱是乌头碱的主要水解产物.理论预测与实验结果相一致,因此Am1半经验量子化学研究乌头碱类大分子化合物是可行的.

Abstract:Objectve: To study the hydrolyzation mechanism of aconitine, and to explain the detoxicating mechanism of Fuzi by the prolonged decoction. Method: The hydrolyzation mechanism of aconitine was studied by using Am1 of quantum chemistry, and HPLC, LC-MS of analytical chemistry. Result: Benzoylaconitine is forecasted to produce easily theoretically, which is proved by the experimental methods. Conclusion: Benzoylaconitine is the main product of aconitine hydrolyzation, which is come from the theoretical and experimental study. So that the method of quantum chemistry is suitable to the structure.
