摘    要:目的:应用近红外光谱技术建立一种川芎中阿魏酸及藁本内酯的快速测定方法。方法:以HPLC法测定的样品中阿魏酸及藁本内酯的含量为真实值,运用近红外漫反射光谱技术采集50份川芎样品的近红外漫反射光谱,结合偏最小二乘法(PLS)建立阿魏酸及藁本内酯含量的定量分析模型。结果:所建阿魏酸及藁本内酯校正模型的相关系数(R2)和内部交叉验证均方差(RMSECV)分别为0.9833,0.0175和0.9771,0.1550;经外部验证,模型的预测相关系数(r2)和预测均方差(RMSEP)分别为0.9819,0.0095和0.9335,0.1139。结论:该方法简便,快速,无污染,可用于川芎中阿魏酸和藁本内酯含量的快速测定。

Rapid Determination of Ferulic Acid and Ligustilide in chuanxiong by Near-infrared Spectroscopy Technique

Abstract:Objective: To establish a rapid analytical method for Ferulic Acid and Ligustilide in chuanxiong by near-infrared diffuse reflectance technique. Methods: HPLC method was used to determine the content of Ferulic Acid and Ligustilide in samples. At the same time, NIR diffuse reflectance spectra of 50 samples were collected. Then, Partial Least-Squares (PIS) method was used to create a quantitative model of calibration of Ferulic Acid and Ligustilide. Results: The correlation coefficients of the PLS calibration models of NIR spectroscopy for Ferulic Acid and Ligustilide were 0. 9833 and 0. 9771, the root-mean-square error of cross-validation (RMSECV) were 0. 0175 and 0. 1550; the correlation between the reference value of the testing sample set and the near infrared predictive value were 0. 9819 and 0. 9335 ; the root-mean-square error of prediction (RMSEP) was 0. 0095 and 0. 1139, Conclusion: The method is simple and convenient, rapid and nondestructive, which can be applied to the fast determination of Ferulic Acid and Ligustilide in chuanxiong
