紫衫醇与顺铂诱导卵巢癌SKOV_3细胞凋亡的研究

摘    要:目的 :研究两种不同的抗癌药物诱导细胞凋亡的规律 ,为卵巢癌的化疗提供理论依据。方法 :紫衫醇与顺铂分别作用于体外培养的卵巢癌SKOV3 细胞系 ,通过组织细胞化学染色、荧光染色、电子显微镜、DNA凝胶电泳、流式细胞术等方法进行检测和观察。结果 :经两种抗癌药物处理的卵巢癌SKOV3 细胞出现典型的细胞凋亡特征 ,在顺铂处理 0~ 36小时内 ,细胞凋亡呈均一的持续性增强 ,在 5~ 2 5nmol ml剂量范围内呈依赖性增强 ,凋亡率最高达 72 %。紫衫醇诱导细胞凋亡亦具有时间和剂量依赖性 ,2 5nmol ml浓度处理 12小时 ,凋亡率最高达 5 5 % ;但时间过长 (>12小时 )或浓度过大 (>5 0nmol ml) ,凋亡率下降。结论 :紫衫醇与顺铂能有效地诱导肿瘤细胞凋亡 ,诱导细胞凋亡是化疗药物治疗癌症的重要机制之一

Abstract:Objective:To observe and compare the pattern of apoptosis induced by anticancer drugs and hence provide a proof for the chemotherapy of ovarian cancer.Methods:Ovarian cancer cell line SKOV 3 was affected by paclitaxel and cisplatin in vitro. Apoptosis was investigated by using cytohistochemistry stain, florescence stain electron microscope scanning, and DNA gel electrophoresis and flow cytometry analysis.Results:Ovarian cancer SKOV 3 cell showed typical characteristic of cell apoptosis after being treated by two anticancer drugs. Apoptosis was enhanced within tested dose range in a dependent pattern. The maximum rate of apoptosis reaches 77%.Apoptosis was 55% in 25nmol/ml in 12 hours. The rate of apoptosis decreased when the time of exposure was longer than 12 hours or the dose is as high as 50nmol/ml.Conclusions:Paclitaxel and cisplatin were effective on inducing ovarian cancer cell. Anticancer drugs induced apoptosis is one of the important mechanism of chemotherapy.
