苦参素穴位注射对肝纤维化大鼠Ⅳ型胶原和TGF-β_1表达的影响

摘    要:目的:观察苦参素足三里(OMZSL)注射对肝纤维化大鼠Ⅳ型胶原和转化生长因子-β1(TGF-β1)表达的影响。方法:用CCl4诱导大鼠肝纤维化模型,随机分为5组:正常组、模型组、苦参素腹腔注射(OMip)组和OMZSL注射治疗组、OMZSL注射预防组。实验至10周末,取肝组织,VG染色观察肝组织中总胶原含量;免疫组化观察Ⅳ型胶原和TGF-β1的表达。结果:与模型组比较OMZSL预防与治疗给药均能明显降低肝纤维化大鼠肝组织中总胶原含量,减少Ⅳ型胶原、细胞因子TGF-β1的表达。结论:OMZSL给药能明显减少肝纤维化组织中总胶原含量及Ⅳ型胶原、TGF-β1的表达,抑制细胞外基质沉积,从而减轻或逆转肝纤维化。

Abstract:Objective:To observe the effects of acupoint injection with oxymatrine on expression of type Ⅳ collagen and transforming growth factor β1(TGF-β1)mRNA on rat liver fibrosis .Methods:The liver fibrosis model in rats was established by injecting carbon tetrachloride(CCl4). Thirty-five SD rats were randomly divided into the normal control group, the model control group, the control group of oxymatrine intraperitoneal injection, the treatment group of oxymatrine injection into zusanli (ZSL) and the preventive group of oxymatrine into ZSL. At the end of the tenth week, degrees of fibrosis were assessed with Van Gieson staining. The expressions of type Ⅳ collagen and TGF-β_1 mRNA were observed by immunohistochemistry. Results:In the treatment and preventive groups of oxymatrine into ZSL, the content of total collagen in liver fibrosis tissue was reduced. In the treatment and preventive groups of oxymatrine into ZSL, degrees of fibrogenesis in liver tissues were markedly reduced compared with the model group. The pathological studies showed that the liver fibrosis degree of oxymatrine ZSL preventive group was lighter than that of the treating group. Expressions of type Ⅳ collagen and TGF-β_1 mRNA in the liver tissue were reduced in the oxymatrine ZSL preventive group and the therapeutic group. Conclusions:In the treatment and preventive groups of oxymatrine injecion into ZSL, the content of total collagen was reduced on the liver fibrosis of rats induced by CCl_4 ,the antifibrotic mechanism was partly improved through reducing expression of type Ⅳ collagen and TGF-β_1 mRNA in the liver tissue, facilitating degradation of the extracellular matrix and preventing ECM deposition in liver.
