| 摘 要： | 目的:运用考马斯亮兰G250显色剂测定壳聚糖中蛋白含量。方法:蛋白质分子均具有酰胺基团,考马斯亮兰G250染料上的阴离子与蛋白质上的酰胺结合,使溶液变为蓝色,于595nm处测定吸光度可计算出样品中蛋白质的含量。结果:考马斯亮兰G250分光光度法测定壳聚糖样品中蛋白质含量是可行的,相对标准偏差为3.6%～4.9%,回收率为93.2%～108.7%。结论:该法具有简便、快速、灵敏度高、重现性好等特点。

| Abstract: | Objective:To determination of concentration of protein in chitosan with coomassie breiliant blue G-250.Methods:All protein contain acylamide group.When the coomassie breiliant blue G-250 encoumter protein solution,its anion will bind acylamide group,and the solution turn blue from red.So the concentration of protein can be determined at 595 nm.Results:Its relative standard deviation is 3.6%~4.9%,and its recovery ration is 93.2%~108.7%.Conclusion:This assay can be as simple,rapid,reproducibility and sensitive method for protein quantities in chitosan. |
