| 摘 要： | 细胞周期是由周期依赖激酶(CDK)和细胞周期蛋白共同作用而完成。尤其是G1期过渡到S期受细胞周期负调节因子家族周期蛋白依赖激酶抑制因子(CDKI)的调节,p21可以和p53共同构成细胞周期G1检查站,因DNA损伤后不经过修复则无法通过,减少受损DNA的复制和积累,从而发挥抑癌作用。当p21、Cyclin、CDK,PCNA四聚体的功能受到抑制,则使增殖信号大量活化,导致细胞异化和恶变。p21作为一种负性调节因子,在依赖野生型p53的表达通路上与肿瘤的表达、表达含量、表达位置,以及预后都密切相关。
