药物性肝损伤机制

摘    要:肝脏是药物在体内代谢的最主要场所,是最容易遭受药物影响的脏器。由于各种药物的广泛应用,药源性肝损伤已经成为严重的健康问题,深入研究其机制,对药源性肝损伤的预防和治疗有很大的帮助。药源性肝损伤机制涉及诸多方面,文章就近些年来研究比较透彻和并得到公认的药源性肝损伤机制如钙平衡破坏和细胞膜损伤、胆汁淤积和胆小管损伤、细胞色素P450代谢激活、自身免疫反应、细胞凋亡以及线粒体损伤等六方面进行了综述。

Abstract:Liver is the major organ that is responsible for the metabolism and elimination of many substances.Toxic injury occurs in the liver mere often than that in any other organ.When drug is used widely, drug-induced liver injury has become a serious health problem in contemporary society,then research on the mechanism of drug-induced liver injury is very useful in therapy and prevention of dreg-induocd liver injury. In this article, the author reviewed recent advances in mechanism of drug-induced liver injury including disroption of calcium homeostasis and cell membrane injury, canalicular and cholestatic injury, metabolic bioactivafion by cytochrome P450 enzymes, antoimmunity, apoptosis and mitocliondrial injury.
