基质金属蛋白酶及其抑制剂与ALI/ARDS的研究进展

摘    要:急性肺损伤(ALI)是以弥漫性肺泡-毛细血管膜损伤所致肺水肿和微肺不张为病理特征的临床危重症,重度ALI即为急性呼吸窘迫综合征(ARDS)。基质金属蛋白酶(MMPs)可破坏肺泡-毛细血管基膜和细胞外基质(ECM),同时MMPs与MMPs抑制剂的不平衡状态引起ECM组成部分的降解,最终导致ALI/ARDS。而MMPs抑制剂可以减轻肺损伤程度,因此有必要开发出特异性高、不良反应小的MMPs抑制剂,为ALI/ARDS的防治提供新的思路。

Abstract:Acute lung injury(ALI) is characterized by pulmonary edema and atelectasis caused by diffuse alveolar-capillary membrane injury.The severe ALI is acute respiratory distress syndrome(ARDS).Matrix metalloproteinases(MMPs) could destroy alveolar-capillary membrane and extracellular matrix(ECM),and the unbalance between MMPs and MMPs inhibitors could induce the degradation ECM,which results in ALI/ARDS finally.So inhibitors of MMPs can reduce the degree of lung injury.It is necessary to develop MMPs inhibitors of high specificity and low toxicity,which can provide new methods to prevent and treat ALI/ARDS.
