摘    要:Toll样受体(TLR)是一类天然免疫受体,最早发现其与果蝇胚胎背、腹侧的发育有关。随着研究的深入,哺乳动物体内也发现有类似同源性受体,并统一称TLR家族。TLR的分布十分广泛,可以识别某些病原体或其产物所共有的特定结构。TLR4是介导内毒素/脂多糖应答的主要受体,而TLR4/CD14信号通路是介导内毒素诱导的炎性反应的重要通路。营养性肥胖、动脉粥样硬化、心肌梗死、哮喘等所引起的非细菌性炎性反应也与TLR4相关。本文就TLR4信号通路与炎性反应的研究进展进行综述。

Abstract:Tall like receptors (TLR)are innate immune receptors related to the proteins in Drosophila regulating dorsal ventral patterning during embryogenesis. To date, the TLR family has also been found in mammalian cells. Toll like receptors distribute widely, whcih present on the cell surface of innate immune cells and have recognize microbial motifs, named as pathogen-associated molecular patterns(PAMPs). TLR4 is the major receptor recognizing various bacterial cell wall components including lipopolysaccharide (LPS) response and signaling of human monocytes upon LPS from gram-negative bacteria depends on the presence and function of both CD14 and TLR4, so the TLR4/CD14 signal pathway is the most important way in inflammation reaction inducing by LPS Variation in innate immunity genes such as TLR4 ,or CD14 may modulate responsiveness to LPS and other PAMPs and thus play a role in the development of atopyand respiratory disease. TLR4 is associated with alimentary obesity, atherosclerosis, cardiac infarction, asthma. This article reviewed the development between TLR4 and inflammation reaction.
