大鼠蛛网膜下腔出血脑基底动脉内皮素受体表达的变化

摘    要:目的:探讨实验性蛛网膜下腔出血诱发脑血管痉挛时内皮素受体-A(ETA)基因在大鼠脑基底动脉的表达变化。方法:36只Wistar大鼠随机分为正常组、蛛网膜下腔出血组(3、7、14d)、生理盐水组和假手术组,HE染色观察大脑基底动脉的病理改变,应用逆转录-聚合酶链反应(RT-PCR)检测各组实验动物脑基底动脉ETAmRNA的表达变化。结果:蛛网膜下腔出血组脑基底动脉明显痉挛;蛛网膜下腔出血后3、7d脑基底动脉ETAmRNA表达较正常组明显增高,14d组趋于正常。结论:内皮素受体-A在蛛网膜下腔出血诱发脑血管痉挛的病理生理过程中可能起重要作用。

Abstract:Objective :To detect the changes of Endothelin-A receptor mRNA in the cerebral basilar artery of rats with experimental subarachnoid hemorrhage induced cerebral vessel spasm. Methods :Thirty-six Wistar rats were randomly divided into normal, experimental, sham-operated and saline-control group. The experimental group was subdivided into subarachnoid hemorrhage (SAH)3, 7 and 14d group. RT-PCR was used to study the changes of Endothelin A receptor mRNA level in the cerebral basilar artery of rats with experimental subarachnoid hemorrhage-induced cerebral vessel. Results ..ET-A receptor mRNA level of basilar artery in SAH 3d and 7d group was higher obviously than that in normal group, sham-operated and saline- control groups. ET-A receptor mRNA cerel in 14d group was normal. Conclusion :ET-A receptor may play an important role in pathogenesis of SAH2induced cerebral vessel spasm.
